摘要
目的建立外周血lnc-HOTAIRM1检测的实时荧光定量PCR(RT-qPCR)方法,并探讨其在APL治疗中的临床应用价值。方法通过ATRA诱导NB4细胞株分析lnc-HOTAIRM1表达规律。优化RT-qPCR的扩增体系和扩增条件,建立外周血lnc-HOTAIRM1 RT-qPCR检测方法。最后评价外周血lnc-HOTAIRM1检测在APL治疗监测中的临床应用价值。结果成功优化得到lnc-HOTAIRM1 RT-qPCR扩增体系和扩增条件。NB4细胞株随着ATRA诱导分化时间的增加,lnc-HOTAIRM1的相对表达量逐渐增加(P<0.05)。在临床外周血标本RT-qPCR检测中,APL组lnc-HOTAIRM1的相对表达量低于非APL组和正常对照(P<0.01);与治疗前APL比较,APL治疗后的lnc-HOTAIRM1相对表达量明显上调(P<0.01)。结论APL组、非APL组和正常对照之间的外周血lnc-HOTAIRM1存在差异表达,lnc-HOTAIRM1与APL诱导分化相关,APL治疗后的lnc-HOTAIRM1表达明显上调。本研究所建立的lnc-HOTAIRM1 RT-qPCR检测方法有望应用于APL患者的鉴别诊断、疗效评估和MRD监测。
Objective To establish a real-time quantitative PCR(RT-qPCR)method for the quantitative peripheral blood(PB)lnc-HOTAIRM1,and to explore its clinical application value in the treatment of APL.Methods The expression of NB4 cell line induced by ATRA was analyzed.The amplification system and amplification conditions of RT-qPCR were optimized,and the PB lnc-HOTAIRM1 RT-qPCR detection method was established.Finally,the clinical value of PB lnc-HOTAIRM1 detection in monitoring APL treatment was evaluated.Results The lnc-HOTAIREM1 RT-qPCR amplification system and amplification conditions were successfully optimized.The relative expression of lnc-HOTAIREM1 increased gradually with the increase of ATRA-induced differentiation time in NB4 cell line(P<0.05).In RT-qPCR detection for clinical PB samples,the relative expression level of lnc-HOTAIREM1 in APL group was lower than that in non-APL group and normal control group(P<0.01).Compared with APL before treatment,the relative expression of lnc-HOTAIREM1 in APL after treatment was significantly up-regulated(P<0.01).Conclusion There were differences in the expression of PB lnc-HOTAIREM1 among APL group,non-APL group and normal control group.lnc-HOTAIREM1 was associated with APL induced differentiation,and the expression of lnc-HOTAIREM1 in APL after treatment was significantly up-regulated.The established RT-qPCR for the quantitative detection of PB lnc-HOTAIREM1 can be used for the differential diagnosis and minimal residual leukemia(MRD)assessment for APL patients.
