摘要
目的探讨加味大柴胡汤对梗阻性黄疸大鼠肝损伤及JNK、Bcl-2 mRNA和蛋白表达的影响。方法将90只SD雄性大鼠按随机数表法分为假手术组(S组)、梗阻性黄疸组(O组)及干预组(M组),每组30只。S组与O组每日定时生理盐水10μL/g灌胃,M组以生药量1 g/mL的加味大柴胡汤10μL/g灌胃,各组按时间点分别于术后3 d、7 d、10 d取材,每次每组分别随机取10只大鼠。检测各组血清天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、血清总胆红素(TBIL)水平,实时荧光定量PCR(qPCR)和Western blot分别检测肝组织JNK、Bcl-2的mRNA及蛋白表达情况。结果O组与M组血清AST、ALT、TBIL均较S组升高,且M组均低于O组(P<0.05)。O组、M组JNK的mRNA及蛋白水平均高于S组,且M组均低于O组(P<0.05);O组、M组Bcl-2的mRNA及蛋白水平均低于S组(P<0.05),且M组均高于O组(P<0.05)。结论加味大柴胡汤能够改善梗阻性黄疸大鼠肝损伤,促进肝组织修复,这可能与下调JNK蛋白表达,促进Bcl-2蛋白表达,从而减弱内质网应激,抑制肝细胞的凋亡有关。
Objective To investigate the effects of Jiawei Dachaihu decoction on liver injury and expressions of JNKand Bcl-2 in obstructive jaundice rats.Methods Ninety SD male rats were divided into sham operation group(group S),terrieryellow group(group O)and intervention group(group M)according to random number table method,30 rats in each group.GroupS and group O were gavaged with 10μL/g of normal saline,group M was gavaged with 10μL/g of Jiawei Dachaihu decoction witha crude drug amount of 1 g/mL.Each group was sampled at 3 d,7 d and 10 d after the operation,and 10 rats were randomly taken per group at each point.The serum levels of aspartate aminotrans ferase(AST),alanine aminotransferase(ALT)and totalbiliary red(TBIL)were detected in the three groups.RT-PCR and Western blot assay were used to detect m RNA and proteinexpressions of liver tissue JNK and Bcl-2,respectively.ResultsThe serum levels of AST,ALT and TBIL were significantly higher in group O and group M than those in group S,and which were significantly lower in group M than those of group O(P<0.05).The levels of JNK m RNA and protein were significantly higher in group O and group M than those in group S,and whichwere significantly lower in group M than those in group O(P<0.05).The levels of Bcl-2 m RNA and protein were significantly lower in group O and group M than those of group S(P<0.05),and which were significantly higher in group M than those of groupO(P<0.05).Conclusion Jiawei Dachaihu decoction can improve liver injury and promote liver tissue repairing in rats with obstructive jaundice,which may be related to the down-regulation of JNK protein,the increase of the expression level of Bcl-2 protein,the weakening of the stress of endoplasmic reticulum and the inhibition of the apoptosis of liver cells.
