摘要
目的研究YWHAZ在主动脉夹层患者主动脉壁组织中的表达,探讨YWHAZ与主动脉夹层发病的关系。方法选取确诊为Stanford A型主动脉夹层患者13例为夹层组,以及无主动脉疾病的心脏移植受体11例为对照组,通过H&E染色及EVG弹力纤维染色比较其形态学差异,通过免疫组化染色对比两组主动脉壁组织中YWHAZ的表达差异及亚细胞定位情况,最后通过提取两组冻存主动脉壁组织中的总蛋白进行蛋白质免疫印迹(Western blot)检测YWHAZ蛋白表达差异。结果H&E及EVG染色结果显示,与对照组相比夹层组患者主动脉壁中平滑肌细胞排列紊乱,弹力纤维断裂不连续。免疫组化染色YWHAZ阳性面积定量分析结果提示夹层组为4.54±0.59,对照组为0.58±0.43,夹层组与对照组比较YWHAZ水平明显上调,具有统计学差异(P<0.01)。Western blot显示YWHAZ蛋白定量结果为对照组1.0±0.16,夹层组1.24±0.31,两组差异具有统计学差异(P<0.05),趋势与免疫组化一致。结论YWHAZ的高表达可能会促进主动脉夹层的发生发展,有望成为治疗主动脉夹层的潜在靶点。
Objective To study the expression of YWHAZ in aortic wall in patients with aortic dissection,and discuss the relationship between YWHAZ and aortic dissection.Methods The patients with diagnosed Stanford A type aortic dissection(n=13)were chosen into aortic dissection group and patients received heart transplantation without aortic disease(n=11)were chosen into control group.The difference in morphology was compared after HE staining and EVG staining,and YWHAZ expression in aortic wall and cellular distribution were compared by using immunohistochemical staining(IHC)in 2 groups.The protein expression of YWHAZ was detected by using Western blotting assay after extracting the total protein cryopreserved in aortic wall.Results The results of HE staining and EVG staining showed that disarranged smooth muscle cells and fragmented elastic fibers in aortic dissection group compared with control group.The results of IHC quantitative analysis showed that positive area of YWHAZ was(4.54±0.59)in aortic dissection group and(0.58±0.43)in control group(P<0.01).The results of Western blotting assay showed that YWHAZ protein quantity was(1.0±0.16)in control group and(1.24±0.31)in aortic dissection group(P<0.05),which showed the same trend as the results of IHC.Conclusion The higher expression of YWHAZ may improve the generation and development of aortic dissection,and it is expected as a potential target in aortic dissection treatment.
