铝灰渣提取物对大鼠消化系统的毒性作用

摘要 为探索铝灰渣提取物(AAE)对大鼠消化系统的毒性机制,将64只SPF级Wistar大鼠按性别体重随机分为4组,设AAE高、中、低剂量组(染毒剂量分别为15.6,7.8,5.2 mg·kg^-1)和对照组(超纯水),连续灌胃14 d.观察各组大鼠一般状态,检测血清TNF-α和NO含量、胃肠组织DAO含量和MPO活性,观察大鼠胃肠平滑肌细胞的组织形态.结果显示,与对照组比较,灌胃第7 d和第14 d,AAE各剂量组大鼠2 h内排便量明显增多,均出现稀便、不定形软便;AAE各剂量组大鼠血清TNF-α和NO含量显著增高,胃和结肠组织DAO含量和MPO活性均显著增高,呈剂量依赖性;AAE对大鼠胃肠黏膜和胃肠平滑肌细胞组织形态学影响大,表现为胃肠黏膜溃疡,萎缩和炎性细胞浸润.研究显示灌胃AAE对大鼠消化系统有一定的毒性作用. To explore the toxicity mechanism of aluminum ash extract(AAE)on digestive system for rats,sixty-four SPF Wistar rats was randomly divided into four groups.AAE were given to the rats of each group with the different dosages(15.6,7.8,5.2 mg·kg^-1,respectively),and the distilled water was given to control group,each group was given by intragastric administration for 14 d.The general condition of rats,TNF-α,NO,DAO,MPO and the histomorphology of gastrointestinal smooth muscle cells were investigated.The results showed that compared with the control group at 7 d and 14 d of gavage,all of the rats with AAE feeding show the increased frequency of thinning defecation within 2 h,the increased TNF-αand NO in serum,and the increased activity of DAO and MPO in gastric and colon tissues,dose-dependently.Besides that,gastrointestinal mucosa ulcer,atrophy and inflammatory cell infiltration in digestive tissues are revealed in the rats with AAE.It is indicated that AAE can induce the toxic effect on the digestive system in rats.
