Rapid immunological detection of olaquindox residues in feed

摘要 基于间接竞争酶联免疫吸附测定(ic-ELISA)方法,建立了一种快捷、灵敏、准确的检测饲料中喹乙醇残留的方法.结果显示:以猪饲料、鸡饲料、鱼饲料为样品原材料,最佳反应条件下得到的标准曲线检测限为(0.6±0.2)μg/L,灵敏度为(3.2±0.3)μg/L,在0.9~8.0μg/L质量浓度范围内线性良好.3种饲料样品中的喹乙醇在低、中、高3个加标浓度水平下的添加回收率为88.7%~100.0%,变异系数为2.3%~11.1%,其结果与高效液相色谱法(HPLC)一致;市售18个饲料样品中,喹乙醇的检出率为50.0%.利用HPLC法对ic-ELISA的结果进行验证,结果一致. Based on the indirect competitive enzyme-linked immunosorbent assay(ic-ELISA)method,a rapid,sensitive and accurate method for the detection of olaquindox residues in feed was established.The suitability of the ELISA for olaquindox quan-tification in pig feed,chicken feed and fish feed was also studied.Under the optimized conditions,the detection is limited by the obtained calibration curve:(0.6±0.2)μg/L,and the sensitivity is(3.2±0.3)μg/L.The good linear responses were obtained in the mass concentration range of 0.9-8.0μg/L.The recovery of olaquindox in three feed samples at low,medium and high levels of spiked concentration was studied.The recovery of olaquindox from spiked samples ranged from 88.7%-100.0%,and the coefficient of variation was 2.3%-11.1%,which is in accordance with the results obtained by the method of HPLC.18 samples of feed purchased from various markets were tested,and the detection rate of olaquindox was 50.0%.Validation of the ic-ELISA was conducted by comparison of results from HPLC,and the results showed consistency.
