姜黄素对VEGF诱导下HaCaT细胞增殖及miR-203表达的影响 被引量:3

摘要 目的观察姜黄素对血管内皮生长因子(VEGF)干预后Ha Ca T细胞增殖及细胞内微小RNA-203(mi R-203)表达的影响。方法 CCK-8法检测不同浓度姜黄素(0、19.5、39、78.1、156.2、312.5、625、1250、2500、5000ng/m L)对VEGF刺激干预后Ha Ca T细胞增殖的影响。实时荧光定量PCR(Real-time PCR)法检测25ng/m L VEGF、1380ng/m L姜黄素、5000ng/m L姜黄素分别及联合作用下对Ha Ca T细胞内mi R-203表达的影响。结果姜黄素在实验浓度范围内呈浓度依赖性抑制Ha Ca T细胞增殖,与0ng/m L姜黄素比较,2500ng/m L姜黄素明显抑制细胞增殖[(0.383±0.006)比(0.520±0.017),P<0.05]。与空白组比较,25ng/m L VEGF诱导下的Ha Ca T细胞mi R-203表达明显上调[(4.049±0.302)比1,P<0.05],而5000ng/m L姜黄素能明显抑制Ha Ca T细胞mi R-203表达[(0.449±0.052)比1,P<0.05],下调25ng/m L VEGF刺激干预下Ha Ca T细胞mi R-203表达[(1.737±0.130)比(4.049±0.302),P<0.05]。结论姜黄素可能通过下调mi R-203表达抑制Ha Ca T细胞过度增殖。 Objective To observe the effect of curcumin on HaCaT cell proliferation and miR-203 expression after intervention of vascular endothelial growth factor(VEGF).Methods CCK-8 method was used to detect the effects of different concentrations of curcumin(0,19.5,39,78.1,156.2,312.5,625,1250,2500 and 5000ng/mL,respectively)on the proliferation of HaCaT cells after VEGF stimulation.Real-time PCR was used to examine the effect of 25ng/mL VEGF,1380 and 5000ng/mL curcumin on the expression of miR-203 in HaCaT cells,respectively and in combination.Results Curcumin inhibited the proliferation of HaCaT cells in a concentration-dependent manner within the experimental concentration range.Compared to 0ng/mL curcumin,2500ng/mL curcumin significantly inhibited cell proliferation[(0.383±0.006)vs(0.520±0.017),P<0.05].Compared to the blank group,the expression of miR-203 in HaCaT cells induced by 25ng/mL VEGF was significantly up-regulated[(4.049±0.302)vs 1,P<0.05],while 5000ng/mL curcumin significantly inhibited the expression of miR-203[(0.449±0.052)vs 1,P<0.05],and down-regulated the expression of miR-203 under 25ng/mL VEGF stimulation intervention[(1.737±0.130)vs(4.049±0.302),P<0.05].Conclusion Curcumin may inhibit HaCaT cell overproliferation by down-regulating MiR-203 expression.
