摘要
目的探讨脂质运载蛋白2(lipocalin 2,Lcn2)在结直肠癌中的表达及其与临床病理特征的关系。方法收集40例配对结直肠癌及癌旁组织样本,分别采用qRT-PCR法、Western blot法检测Lcn2的mRNA和蛋白水平,运用免疫组化检测Lcn2蛋白并分析其与临床病理特征的关系。结果与癌旁组织相比,结直肠癌组织中:qRT-PCR结果显示Lcn2的mRNA水平升高(P=0.0065);Western blot结果显示Lcn2的蛋白水平升高(P<0.05);免疫组化结果显示,癌细胞胞质和胞膜中Lcn2呈强阳性,癌旁组织Lcn2呈阴性,且Lcn2强阳性与肿瘤直径(P=0.014)、分化程度(P=0.017)、T分期(P=0.042)及淋巴结转移(P=0.025)密切相关。结论Lcn2在结直肠癌中呈强阳性,且与肿瘤直径、分化程度、T分期及淋巴结转移密切相关,提示其可能参与结直肠癌的进展和转移。
Purpose To investigate the expression of lipocalin 2(Lcn2)in colorectal cancer and to analyze its relationship with clinicopathological features.Methods Fourty paired colorectal cancer and paracancerous tissues were collected.The transcriptional mRNA levels and protein expression of Lcn2 were detected by qRT-PCR and Western blot,respectively.And immunohistochemical staining was used to detect Lcn2,the results of Lcn2 and its relationship with clinicopathological features were analyzed.Results Compared with the normal tissues,the results of qRT-PCR(P=0.0065)and Western blot(P<0.05)showed that Lcn2 was significantly overexpressed in colorectal cancer tissues.And immunohistochemical study also indicated that the colorectal cancer cells were positive for Lcn2 in the cytoplasm and membrane,while negative in normal tissues.And its high expression was strongly associated with tumor size(P=0.014),differentiation(P=0.017),T stage(P=0.042)and lymph node metastasis(P=0.025).Conclusion The high expression of Lcn2 in colorectal cancer is strongly associated with clinical tumor size,histopathological differentiation,T stage and lymph node metastasis,which may be involved in the progression and metastasis of colorectal cancer.
