探讨Wnt1、Wnt5a和β-catenin在胃腺癌中表达的临床意义 被引量:1

摘要 目的分析Wnt1、Wnt5a和β-catenin在胃腺癌中表达的临床意义,并探讨三者在胃腺癌中表达的相关性及可能参与胃腺癌发生进展的机制。方法 SABC免疫组化法检测86例胃腺癌组织及与之对应的癌旁组织和正常胃组织中Wnt1、Wnt5a和β-catenin的表达情况。结果 Wnt1、Wnt5a和β-catenin在胃腺癌组织中的表达高于与之对应的癌旁组织,在癌旁组织中的表达高于正常胃组织(P<0.05);Wnt1和β-catenin在胃腺癌组织中的高表达相关于癌灶大小、分化程度、浸润深度、TNM分期、有无淋巴结转移及血管浸润(P<0.05),Wnt5a在胃腺癌组织中的表达与浸润深度、TNM分期、有无淋巴结转移及血管浸润相关(P<0.05)。Wnt1和β-catenin在胃腺癌中的表达无相关性(P>0.05),而Wnt5a和β-catenin的表达呈负相关(r=-1,P<0.01)。β-catenin的表达在胃腺癌组织中主要位于细胞质与细胞核,在正常胃粘膜组织中主要位于细胞膜。结论 Wnt1、Wnt5a和β-catenin在胃腺癌中的表达均升高,三者发挥癌基因样作用促进胃腺癌的发生进展。Wnt1和β-catenin可能通过激活经典Wnt信号通路发挥促癌作用,β-catenin由细胞膜到细胞质再到细胞核的异位表达提示胃腺癌的进展。Wnt5a可能经由其它信号通路在胃腺癌的浸润转移中发挥作用。 Objective To analyze the clinical significance of Wnt1,Wnt5 a and β-catenin expression in gastric adenocarcinoma,and to explore the correlation of three expressions in gastric adenocarcinoma and possible mechanisms involved in the occurrence and progression of gastric adenocarcinoma.Methods SABC immunohistochemical method was used to detect the expression of Wnt1,Wnt5 a and β-catenin in 86 cases of gastric adenocarcinoma tissues and corresponding adjacent tissues and normal gastric tissues.Results The expressions of Wnt1,Wnt5 a and β-catenin in gastric adenocarcinoma tissues were higher than those in corresponding adjacent tissues,and the expressions in adjacent tissues were higher than that in normal gastric tissues( P< 0. 05).The high expressions of Wnt1 and β-catenin in gastric adenocarcinoma tissues were related to the size,degree of differentiation,and infiltration depth,TNM staging,lymph node metastasis and vascular invasion( P<0. 05).The Wnt5 a in gastric adenocarcinoma tissue was related to the depth of invasion,TNM stage,lymph node metastasis and vascular invasion( P<0. 05).There was no correlation between Wnt1 and β-catenin in gastric adenocarcinoma expression( P > 0. 05),while the expressions of Wnt5 a andβ-catenin were negatively correlated( r=-1,P<0. 01).The expression of β-catenin was mainly located in the cytoplasm and nucleus of gastric adenocarcinoma tissues,and in the cell membrane of normal gastric mucosal tissues.Conclusion The expressions of Wnt1,Wnt5 a and β-catenin would be elevated in gastric adenocarcinoma that play an oncogene-like effect to promote the occurrence and progression of gastric adenocarcinoma.Wnt1 and β-catenin may play a role in promoting cancer by activating the canonical Wnt signaling pathway.The ectopic expression of β-catenin from the cell membrane to the cytoplasm to the nucleus would indicate gastric adenocarcinoma progression.Wnt5 a may play a role in the invasion and metastasis of gastric adenocarcinoma via other signaling pathways.
