摘要
目的探究在骨关节炎(osteoarthritis,OA)与类风湿关节炎(rheumatoid arthritis,RA)中的正五聚蛋白3(PTX3)蛋白的表达情况,观测经IL-1β诱导后ADTC5软骨前体细胞PTX3蛋白表达情况并探讨其可能机制,利用siRNA干扰PTX3表达观察软骨细胞相关合成代谢指标表达。方法检测OA患者与RA患者的PTX3的表达;对小鼠进行OA与RA模型建立,对以上标本进行组织学染色,观察软骨,滑膜中PTX3蛋白的表达情况;使用IL-1β诱导ADTC5细胞构建OA炎症细胞模型,观察其PTX3蛋白的表达;敲减PTX3后检测ADTC5相关指标的表达情况。结果PTX3在OA与RA的软骨与滑膜中表达增高;且在利用IL-1β诱导ADTC5后,PTX3蛋白表达增多;使用siRNA敲减PTX3后,Western blot结果显示MMP13表达减少,COL2表达增多。结论PTX3在OA和RA的软骨中表达增多,且可能参与了骨性关节炎的发展。
Objective To explore the role of PTX3 in arthritis.The expression of PTX protein in OA and RA was investigated.The expression of PTX3 protein in ADTC5 cartilage precursor cells induced by IL-1βwas observed and its possible mechanism was determined.The expression of anabolic markers related to chondrocytes was observed by siRNA interference with PTX3 expression.Methods Detection of PTX3 expression in patients with OA and RA.The mice were established with OA and RA models.The above specimens were histologically stained to observe the expression of PTX3 protein in cartilage and synovial membrane.ADTC.5 cells were induced to construct OA inflammatory cell model by IL-1β,and the expression of PTX3 protein was observed.The expression of ADTC5 related indicators was detected after knockdown of PTX3.Results The expression of PTX3 was increased in cartilage and synovium of OA and RA.The expression of PTX3 protein was increased after ADTC5 induced by IL-1β.After knockdown of PTX3 with siRNA,MMP13 expression was decreased and COL2 expression was increased.Conclusion The expression of PTX3 is increased in the cartilage of osteoarthritis and rheumatoid arthritis,and may be involved in the development of osteoarthritis.
