长链非编码RNA TUSC8在宫颈癌中的表达及临床意义

Clinical significance of long non - coding RNA TUSC8 in cervical cancer.

目的检测长链非编码RNA TUSC8(lncRNA tumor suppressor candidate 8)在宫颈癌(cervical cancer,CC)组织中的表达,探讨lncRNA TUSC8与CC临床病理特征及预后间的关系.方法RT-PCR检测CC组织和配对癌旁组织以及CC细胞中lncRNA TUSC8的表达;利用单因素方差分析lncRNA TUSC8与CC患者临床病理特征之间的关系;利用生存曲线分析lncRNA TUSC8与CC患者术后5年生存率的关系;利用Cox风险比例模型分析lncRNA TUSC8与CC患者预后的关系.结果lncRNA TUSC8在CC组织和细胞中均呈低表达;lncRNA TUSC8的表达与CC的TNM分期相关(P<0.05);lncRNA TUSC8低表达的CC患者术后5年生存率低于高表达的患者,lncRNA TUSC8低表达是CC患者预后的独立危险因素.结论lncRNA TUSC8在CC中表达降低,并与CC患者生存及预后密切相关.

Objective To investigate the expression level of long non-coding RNA TUSC8(lncRNA tumor sup-pressor candidate 8)in cervical cancer(CC)and evaluate its prognostic value in CC.Methods CC tissue as well as.adjacent normal tissue from patients diagnosed with CC were harvested in this study.Expression of IncRNA TUSC8 was measured by real-time PCR.ANOVA analysis was performed to evaluate the association between lncRNA TUSC8 and clinical manifestation of CC patients.K-M survival analysis was utilized to determine correlation of CC expression with 5-year survival.Cox's proportional hazards regression models were applied to analyze the correlation between lncRNA TUSC8 and prognosis of CC patients.Results lncRNA TUSC8 was down-regulated in CC cells and tissues.Expression of lncRNA TUSC8 was closely associated with TNM stage of CC(P<0.05).Expression of lncRNA TUSC8 was positively correlated with 5-year survival rate.lncRNA TUSC8 was an independent prognostic risk factor of CC.Conclusion ln-cRNA TUSC8 is down-regulated in CC and associated with survival outcome of CC.
