摘要
为提高西藏地区银白杨组培苗的生根率,筛选出银白杨不定芽诱导生根的最佳培养基,以前期银白杨组培中获得的不定芽为试验材料,设置不同光照周期、蔗糖浓度、琼脂粉含量、不同激素配比及不同pH值等培养条件,筛选出最优生根培养基。结果表明,1/2MS为银白杨的最佳基本培养基,最佳激素为IBA浓度0.4mg/L,最适pH值5.7±0.1;光照12h/d、琼脂6g/L、蔗糖15g/L时银白杨不定芽生根率最高且生长态势良好;综合各影响因子,最终确定银白杨不定芽生根的最佳培养基配方为:1/2MS+IBA0.4mg/L+琼脂6g/L+蔗糖15g/L,pH 5.7±0.1。
In order to improve the rooting rate of tissue culture and discuss the optimal medium for adventitious bud rating of Populus alba in Tibet,the adventitious buds obtained from the tissue culture of P.alba torments were used as experimental materials.On the basic of different light,agar,sugar,the best hormone ratio and the optimal pH,the effect of sucrose on the rooting of adventitious buds were researched.The results showed that 1/2 MS was the best basic medium,IBA0.4 mg/L was the optimal hormone concentration,the optimum pH was 5.7±0.1;The illumination time was 12 h/d,the agar concentration was 6 g/L,and the sucrose concentration was 15 g/L,the adventitious bud of white poplar has high rooting rate and good growth state;The optimal medium formula for the comprehensive rooting of adventitious bud rooting of white poplar was 1/2 MS+IBA0.4 mg/L+agar 6 g/L+sugar15 g/L,pH5.7±0.1.
