苏合香丸治疗蛛网膜下腔出血的网络药理学研究 被引量:6

摘要 目的采用网络药理学的研究方式,探讨具有醒脑开窍作用的苏合香丸治疗蛛网膜下腔出血的分子机制,以指导临床应用。方法利用中药相关数据库搜索出苏合香丸的有效化学成分,依据这些化学成分找到针对中枢神经系统的治疗靶点,并与蛛网膜下腔出血的疾病靶点相匹配,找到治疗蛛网膜下腔出血的关键靶基因,进一步应用Systems Dock完成分子对接验证,最后对其靶点采用GSEA法富集分析。结果根据口服生物利用度、类药性及血脑屏障筛选出苏合香丸的11种有效成分,对应着183个靶点,与蛛网膜下腔出血的382个靶点应用cytoscope软件得到最终关键节点21个,这些节点与蛛网膜下腔出血后的血管痉挛、昏迷患者促进苏醒等作用密切相关。结论本研究揭示了苏合香丸治疗蛛网膜下腔出血的作用机制,体现了中药复方多靶点的治疗优势,为进一步药理学研究提供了实验依据。 Objective The purpose of this study was to explore the therapeutic mechanism of Suhexiang Pills in the treatment of subarachnoid hemorrhage based on the network pharmacology approaches.Methods We listed the chemical compounds of Suhexiang Pills and used these compounds to find out therapeutic targets for the central nervous system.Then,those targets were matched with the disease targets of subarachnoid hemorrhage(SAH)to find out the key targets for the treatment of SAH,and Systems Dock method was applied to the molecular docking verification.Finally,GSEA method was used for the targets enrichment analysis.Results After oral bioavailability(OB),drug-likeness(DL)and blood brain barrior(BBB)were set as the screening criteria,11 potential active components in Suhexiang Pills and 183 targets were identified.A total of 21 key nodes were obtained from 183 targets of 11 components merged with 382 targets of SAH using cytoscope software.And these key nodes were closely related to the vasospasm and recovery of consciousness after SAH.Conclusion In this study,we revealed the potential mechanism of Suhexiang Pills in the treatment of SAH and built a systematical network pharmacology approach,which indicated thattraditional Chinese medicine had advantages of multi-targets therapy of SAH.Our results provided a basis for the future study of Suhexiang Pills.
