‘无为大红袍’荸荠组织培养及快速繁殖 被引量:6

摘要 以安徽‘无为大红袍’荸荠[Eelocharis dulcis (Burm.f.) Trin. ex Hensch ‘Wuwei Dahongpao’]的主芽和侧芽为外植体,建立其离体快速繁殖体系。结果表明,荸荠主芽的最适消毒时间为10 min,最适诱导培养基为MS+3.0 mg·L^-1 6-BA,平均出芽率为85.7%,该培养基中萌芽较粗,绿色,长势较好,生长较快;荸荠侧芽的最适消毒时间为8 min,最适诱导培养基为MS+2.0 mg·L^-1 6-BA,平均出芽率为81.8%,该培养基中萌芽粗壮,青绿色,长势好,生长快速。适合‘无为大红袍’荸荠增殖的培养基为MS+0.8 mg·L^-1 6-BA+0.06 mg·L^-1 TDZ+0.15 mg·L^-1NAA,增殖系数为5.43,叶状茎较粗,长势好。在KNO3:(NH4)2SO4 (物质的量浓度之比=1:4)时,茎粗达到最大值,为1.98 mm。当园土:商品基质(体积比=1:1)时,荸荠组培苗假植成活率最高,达到100%,将假植成活的荸荠苗移栽至大田后,成活率达到100%。 The main buds and lateral buds of Eelocharis dulcis(Burm.f.) Trin. ex Hensch ‘Wuwei Dahongpao’ were used as explants to establish a rapid propagation and regeneration system in vitro. The results showed that the most suitable time for disinfection of the main buds of water chestnut was 10 min, and the most suitable medium for induction was MS+3.0 mg·L^-1 6-BA, with an average germination rate of 85.7%. The buds in this medium were thick, green, with better and fast growth. The most suitable time for disinfection of the lateral buds of water chestnut was 8 min, and the most suitable medium for induction was MS+2.0 mg·L^-1 6-BA, with an average germination rate of 81.8%. The buds in this medium were thick and strong, turquoise grew better and fast. MS+0.8 mg·L^-1 6-BA + 0.06 mg·L^-1 TDZ+0.15 mg·L^-1 NAA was the most suitable medium for the multiplication of water chestnut. The multiplication coefficient was 5.43. The leaf like stem was thicker and grew well. When KNO3:(NH4)2 SO4(1:4), the stem diameter reached the maximum value, which was 1.98 mm. When garden soil: commercial substrate(1:1), the survival rate of tissue culture seedlings of water chestnut was the highest, reaching 100%. After transplanting the seedlings to the field, the survival rate reached 100%.
