人内皮型一氧化氮合酶结构和功能的生物信息学分析 被引量:3

摘要 目的运用生物信息学方法分析内皮型一氧化氮合酶(eNOS)的结构和功能。方法利用生物信息学的各种软件,分析eNOS基因启动子、基因同源性,以及其所编码蛋白的理化性质、信号肽、跨膜结构、二级结构、三级结构、翻译后修饰和蛋白质相互作用。结果eNOS基因有14个启动子,基因同源性分析发现人与猪的eNOS基因序列接近度较高。eNOS蛋白属于亲水的不稳定蛋白,不属于分泌蛋白,且没有信号肽;二级结构以α螺旋和无规则卷曲为主,三级结构稳定可靠;N-糖基化位点和O-糖基化位点各有0和29位,磷酸化的位点有Ser:35,Thr:14,Tyr:4。eNOS蛋白可以与10个蛋白发生作用。结论eNOS具有利于蛋白间相互作用的结构,并可在细胞膜定位,具有传递信息的功能。 Objective To analyze the structure and function of endothelial nitric oxide synthase(eNOS)by bioinformatics.Methods Various softwares of bioinformatics were applied to analysis on promoters of eNOS gene and gene homology of eNOS,and the physical and chemical properties,signal peptide,transmembrane structure,secondary structure,tertiary structure and post-translational modification of proteins encoded by eNOS gene,as well as protein interaction.Results The eNOS gene possessed 14 promoters,and the gene homology analysis showed that sequence proximity of eNOS gene between human and pig was relatively high.The eNOS protein was a hydrophilic and unstable protein without signal peptide,but did not belong to secretory protein;the secondary structure was dominated byα-helix and irregular curl,and the tertiary structure was stable and reliable;there was no N-glycosylation site,whereas there were 29 O-glycosylation sites,and the phosphorylated sites included Ser:35,Thr:14,and Tyr:4.eNOS protein could interact with 10 proteins.Conclusion eNOS possesses a structure contributing to interaction between proteins and can locate in the cell membrane,having a function of information transmission.
