摘要
为建立海带愈伤组织高效诱导体系,对海带愈伤组织诱导过程中不同消毒剂及其时间组合、不同诱导培养基及生长素组合、不同接种外植体部位等因素进行了试验研究。结果表明,使用1.5%KI和双抗(60 mg/L青霉素和100 mg/L环丙沙星)各消毒10 min效果最好,对海带组织的损伤最小,愈伤组织成活率最高。用PESI液体培养基加入2.0 mg/L的激素NAA,愈伤组织的诱导率最高,促分化作用显著。海带表皮组织是最佳诱导外植体,诱导出的愈伤组织分化率最高。
In order to establish a high efficient induction system of Sacharina Japonica callus,the effects of different disinfectants and their time combinations,different induction media and hormone combinations,and different explants on callus induction of S.japonica were studied.The results showed that 1.5%KI and double anti-biotics(60 mg/L Penicillin and 100 mg/L Ciprofloxacin)for 10 minutes had the best disinfection effect and the least damage to S.japonica tissue,meanwhile,had the highest survival rate of Callus.2.0 mg/L NAA was added to PESI liquid medium which the callus induction rate was the highest,and the differentiation-promoting effect was significant.The S.japonica epidermis was the best explant because the filamentous differentiation rate was the highest.
