摘要
基于Sequenom MassARRAY■SNP技术,旨在探究绵羊FLI-1基因g.31369206C>T、g.31365178T>C、g.31369230G>A三个位点在单羔绵羊品种(滩羊、萨福克羊、苏尼特羊和草原型藏羊)和多羔绵羊品种(策勒黑羊、湖羊和小尾寒羊)中的多态性及其与小尾寒羊产羔数的关联。结果表明:g.31369206C>T位点在单、多羔羊群体中均只存在CC、CT两种基因型;g.31365178T>C位点在单、多羔羊群体中均存在CC、CT、TT三种基因型,并且该位点的基因型频率和等位基因频率在单、多羔群体间均达到了差异极显著水平(P<0.01);g.31369230G>A位点在多羔羊群体中只存在GG、GA两种基因型,在单羔羊群体中存在GG、GA、AA三种基因型。g.31369206C>T和g.31369230G>A位点在单、多羔绵羊品种中均表现为低度多态(PIC<0.25);g.31365178T>C位点在湖羊、萨福克羊和滩羊中表现为中度多态(0.25<PIC<0.5),在其余4个品种中均表现为低度多态。g.31369206C>T、g.31365178T>C、g.31369230G>A三个位点在7个绵羊品种中均处于哈代-温伯格平衡状态(P>0.05)。产羔数关联分析表明,FLI-1基因g.31369206C>T位点和g.31365178T>C位点多态性与小尾寒羊前3胎产羔数显著关联(P<0.05)。综上所述,推测FLI-1基因g.31369206C>T位点的T等位基因和g.31365178T>C位点的T等位基因是提高小尾寒羊产羔数的潜在分子标记,g.31369230G>A位点有待于进一步验证。
Based on Sequenom MassARRAY■SNP technology,the aim of the present experiment was to elucidate the polymorphism of g.31369206C>T,g.31365178T>C,g.31369230G>A of FLI-1 gene in monotocous(Tan sheep,Suffolk sheep,Sunite sheep,Prairie Tibetan sheep)and polytocous(Cele black sheep,Hu sheep,Small Tail Han sheep)breeds,and to analyze the association between the loci and the litter size in Small Tail Han sheep.The results showed that the genotypes were found at g.31369206C>T(CC,CT),g.31365178T>C(CC,CT,TT)in all sheep breeds,g.31369230G>A in polytocous(GG,GA)and monotocous(GG,GA,AA)sheep breeds.The genotype frequency and gene frequency of g.31365178T>C were extremely significantly different between polytocous and monotocous sheep breeds(P<0.01).Among 7 sheep breeds,g.31369206C>T and g.31369230G>A were at low polymorphism(PIC<0.25).The g.31365178T>C locus was at moderate polymorphism(0.25<PIC<0.5)in Hu sheep,Suffolk sheep,Tan sheep,it was at low polymorphism in other breeds.g.31369206C>T,g.31365178T>C and g.31369230G>A were in Hardy-Weinberg equilibrium(P>0.05)in 7 sheep breeds.The association analysis indicated that g.31369206C>T,g.31365178T>C from the FLI-1 gene had significant correlation(P<0.05)with litter size of the first three parities of Small Tail Han sheep.In summary,it is speculated that the T allele of g.31369206C>T site and the T allele of g.31365178T>C site of FLI-1 gene may be potential molecular markers for improving litter size in Small Tail Han sheep.
