摘要
目的探讨长链非编码RNA RP1-90G24.1在胃癌中的表达及对胃癌细胞BGC-823侵袭和迁移的作用机制。方法选取胃癌组织及对应的癌旁组织样本各120例,实时荧光定量聚合酶链反应(quantitative real-time polymerase chain reaction,RT-qPCR)检测RNA RP1-90G24.1在胃癌组织及对应的癌旁组织样本中的表达。细胞分为空白对照组、阴性对照组、沉默组;将慢病毒载体RNA RP1-90G24.1-siRNA和RNA RP1-90G24.1-siRNA-NC转染至沉默组、阴性对照组细胞中,空白对照组不做处理。用Transwell小室实验检测各组细胞的迁移和侵袭能力。结果 RNA RP1-90G24.1在胃癌中高表达;癌旁组织、胃癌组织中RNA RP1-90G24.1的相对表达量依次为1.01±0.05,5.35±0.24,差异有统计意义(P<0.05)。空白对照组、阴性对照组、沉默组中细胞的相对迁移率分别为(10.01±0.28)%,(10.00±0.25)%和(2.35±0.12)%,通过matrigel基质胶的数量分别为(1238±86),(1206±107),(247±35)个,沉默组与空白对照组比较,差异有统计意义(P<0.05)。结论 RNA RP1-90G24.1在胃癌中呈高表达,沉默RNA RP1-90G24.1的表达能抑制胃癌细胞BGC823在体外及体内的侵袭及迁移能力。
Objective To investigate the expression of long-chain non-coding RNA RP1-90G24.1 in gastric cancer and its mechanism on invasion and migration of gastric cancer cell line BGC-823. Methods 120 cases of gastric cancer tissue and corresponding paracancerous tissue samples were selected. Quantitative real-time polymerase chain reaction(RT-qPCR) was used to detect the expression of RNA RP1-90G24.1 in gastric cancer tissues and corresponding adjacent tissues samples. Cell experiments were divided into the blank control group, the negative control group, and the silent group;the lentiviral vectors RNA RP1-90G24.1-siRNA and RNA RP1-90G24.1-siRNA-NC were transfected into the cells of the silent group and the negative control group. The blank control group was left untreated. Transwell chamber experiments were used to detect the migration and invasion ability of cells in each group. Results RNA RP1-90G24.1 is highly expressed in gastric cancer.The relative expression levels of RNA RP1-90G24. 1 in adjacent cancer tissues and gastric cancer tissues were 1. 01 ± 0. 05 and 5. 35 ± 0. 24,respectively. Compared between adjacent cancer tissues and gastric cancer tissues,the difference was significant(P < 0. 05). The relative migration rate of cells in the blank control group,negative control group and silent group were(10. 01 ± 0. 28) %,(10. 00 ± 0. 25) % and(2. 35 ± 0. 12) %. and the number of the cell passed matrigel matrigel were(1238 ± 86),(1206 ± 107),(247 ± 35). Compared between the silent group and the blank control group,the differences were significant(P < 0. 05). Conclusion RNA RP1-90G24. 1 is highly expressed in gastric cancer,and silenting the expression of RNA RP1-90G24. 1 can inhibit the invasion and migration of gastric cancer cell line BGC823 in vitro and in vivo.
