苗医“石弱骨虚”大鼠模型复制及几种淫羊藿的干预作用

摘要 目的:复制苗医学说指导下的“石弱骨虚”模型大鼠并探讨几种淫羊藿对模型的作用。方法:在苗医药学说指导下采用手术去除大鼠双侧卵巢+肌肉注射地塞米松磷酸钠+给予2.5%氯化锶水溶液的方法复制“石弱骨虚”模型,采用苗族常用的淫羊藿对模型动物进行干预,以方测证。检测大鼠胫骨和股骨重量;取大鼠血清用酶联免疫试剂盒(ELISA)测定雌二醇(E2)、钙离子(Ca^2+)、无机磷(P)、25羟基维生素D3(25(OH)D3)、1,25二羟基维生素D3(1,25(OH)2D3)、骨钙素(osteocalcin,BGP)、碱性磷酸酶(alkaline phosphatase,ALP)含量。结果:与空白组比较,模型组大鼠胫骨和股骨重量;血清中E2、Ca^2+、P、25(OH)D 3、1,25(OH)2D3、BGP和ALP含量均降低;与模型组比较,不同种类淫羊藿组大鼠胫骨和股骨重量;血清中E2、Ca^2+、P、25(OH)D3、1,25(OH)2D3、BGP和ALP含量出现不同程度的升高。结论:实验所复制的复合模型在一定程度上与苗医理论中“石弱骨虚”的含义相对应,表明“石弱骨虚”模型大鼠复制成功;淫羊藿可通过提高成骨细胞的活性、促进骨形成、增加骨矿化、增加骨量、稳定骨矿化速率、促进血钙、磷平衡、改善骨组织健康状态等方式增加大鼠胫骨和股骨的重量,升高血清中E2、Ca^2+、P、25(OH)D3、1,25(OH)2D 3、BGP和ALP的含量,改善大鼠“石弱骨虚”的状态,也一定程度上阐明了其应用于“石弱骨虚”的药理学基础。 Objective The‘Bone Weakness and Deficiency Rat Model’(BWDRM)was replicated under the theory of Miao Medicine,and several species of Herba Epimedii were evaluated for the pharmacological effects of this model.Methods BWDRM was established as follows:Bilateral ovaries of rats were removed,after that on the fourth day,intramuscular injected with 1mg/kg dexamethasone sodium phosphate twice a week and simultaneously water freely with 2.5%strontium chloride in aqueous solution for two months.On that basis,drug groups were gavage with 3g/kg/d Herba Epimedii extractsper day,but model group and control group were isopyknic saline solution as a contrast.After 4 months,shinbones and thighbones were separated and weighed,serum levels of biochemical indices E2,Ca^2+,P,25(OH)D 3,25(OH)2D 3,BGP,ALP were evaluated.Results Lower levels of shinbones and thighbones weight and serum biochemical indices were detected vs normal group and control group.But on the contrast,higher levels were tested in Herba Epimediis groups vs model group.Conclusions BWDRM was carried out on the basis of a complex model with ovariectomied model and drug model,which corresponded with the theory of‘Bone Weakness and Deficiency’(BWD)in Miao Medicine to some extent.Furthermore in this study,Herba Epimediis were evaluated to improve the state of BWD from enhancing the activity of osteoblasts,promoting bone formation,increasing bone mineralization,increasing bone mass,stabilizing bone mineralization rate,promoting the blood calcium,keeping the phosphorus balance,improving bone tissue health to increase weights of shinbones and thighbones and levels of serum biochemical indices E2,Ca^2+,P,25(OH)D3,25(OH)2D3,BGP.
