摘要
NmpC是沙门菌编码的外膜蛋白,也是沙门菌诱导体液免疫的主要抗原。为研究沙门菌NmpC蛋白的免疫特性,本试验对沙门菌nmpC基因进行扩增,并将其克隆到pET-28a载体上,利用大肠杆菌表达系统表达重组蛋白rNmpC,进一步将rNmpC免疫小鼠,检测特异性抗体水平;用肠炎沙门菌C50336菌株感染rNmpC免疫小鼠评测其免疫保护效果。结果显示,成功表达和纯化出肠炎沙门菌NmpC重组蛋白,rNmpC大小为38000;将rNmpC蛋白免疫小鼠,可以刺激小鼠产生高水平特异性抗体;细菌感染试验表明,rNmpC蛋白免疫小鼠可使其产生针对肠炎沙门菌感染的保护力,保护率为70%。本试验证实沙门菌NmpC蛋白的免疫潜力,为进一步研制沙门菌新型疫苗提供了参考。
NmpC is the outer membrane protein encoded by Salmonella,and is also the main antigen of Salmonella inducing humoral immunity.To study the immune characteristics of Salmonella NmpC protein in this study,Salmonella nmpC gene was amplified and cloned into pET-28 a vector.The recombinant protein rNmpC was expressed in E.coli expression system.Then rNmpC was further immunized to mice to detect the specific antibody level.The immune protective effect of Salmonella enteritidis C50336 strain on rNmpC immunized mice was evaluated.The results showed that the recombinant protein of Salmonella enteritidis NmpC was successfully expressed and purified,and the size of rNmpC was 38000.Immunization of mice with rNmpC protein could stimulate mice to produce high level of specific antibodies.The bacterial infection test showed that the recombinant protein of Salmonella enteritidis NmpC was highly specific.Mice immunized with rNmpC protein had protective effect against Salmonella enteritidis infection with a protective rate of 70%.This study confirmed the immune potential of Salmonella NmpC protein.It provides a reference for further development of new Salmonella vaccine.
