五味子醇甲对去甲肾上腺素诱导心肌肥大损伤的cT-I、cT-T、ET-1调控作用的影响 被引量:4

Effects of schisandrin on cT-I,cT-T and ET-1 regulation of norepinephrine-induced myocardial hypertrophy

摘要 培养大鼠源H9C2心肌细胞进行试验,共分为6组,对照组(加入细胞培养液);模型组(10^-6mol/L去甲肾上腺干预48 h);卡托普利组与模型组同样处理后,加入卡托普利5 mg/L;五味子醇甲低、中、高剂量组与模型组同样处理后,分别加入五味子醇甲5,10,20 mg/L,作用48 h。通过观察HE染色细胞的形态变化,图像分析系统测定心肌细胞表面积,MTT法检测细胞存活率,Hoechst33342染色法检测心肌细胞的凋亡率,JC-1法测定线粒体膜电位,Western blot检测CT-I、CT-T、ET-1蛋白表达。结果显示,五味子醇甲可降低心肌细胞凋亡率和提高心肌细胞存活率,对心肌细胞的形态具有改善作用,并下调了CT-1、CT-T、ET-1蛋白表达。结果表明,五味子醇甲对去甲肾上腺素诱导的心肌细胞损伤具有保护作用,可能通过下调CT-I、CT-T、ET-1蛋白表达,降低炎性反应,减少对心肌细胞的损伤凋亡。 To investigate the protective effect of schisandrin on norepinephrine-induced cardiomyocyte hypertrophy,and its effect on the regulatory mechanism of CT-I,CT-T and ET-1.Culture H9 C2 cardiomyocytes for experiments,divided into 6 groups:Control group(add cell culture solution);Model group(10^-6 mol/L norepinephrine intervention for 48 h);After the captopril group,schizandrin low,medium and high dose groups were treated in the same way as the model group,Add captopril 5 mg/L,schisandrin low,medium and high dose groups(schisandrin concentration is 5,10 and 20 mg/L)for 48 h.The morphological changes were observing by HE staine,the image analysis system measures the surface area of myocardial cells,MTT method to detect cell survival rate,Hoechst33342 staining method was used to detect the apoptosis rate of cardiomyocytes,JC-1 method for measuring mitochondrial membrane potential,Western blot was used to detect the protein expression of CT-I,CT-T and ET-1.The results showed that schisandrin can reduce the rate of cardiomyocyte apoptosis and increase the survival rate of cardiomyocytes,improve the morphology of cardiomyocytes,and down-regulate the expression of CT-1,CT-T,ET-1 protein.In conclusion,schisandrin has a protective effect on norepinephrine-induced cardiomyocyte damage,may be by down-regulating the expression of CT-I,CT-T and ET-1 protein,reducing the inflammatory response,reducing apoptosis damage to cardiomyocytes.
