基于LC-MS/MS分析酒炙车前子治疗奶牛胎衣不下活性组分的化学成分 被引量:5

Chemical constituents from active components of semen plantaginis by stir-frying with wine on cure of retained fetal membranes in cows by LC-MS/MS

摘要 采用液相串联质谱法(LC-MS/MS)对酒炙车前子治疗奶牛胎衣不下活性组分Ⅲ和Ⅵ的化学成分进行分析和鉴定。以Hyper gold C18(100.0 mm×2.1 mm,1.9μm)为色谱柱,水(含0.1%甲酸,5%乙腈)-乙腈(含0.1%甲酸)为流动相进行梯度洗脱,流速为0.3 mL/min,柱温为40℃;采用ESI离子源,正、负离子模式下采集活性组分Ⅲ和Ⅵ的一级和二级质谱数据。根据碎片离子与精确相对分子质量信息同Metlin、HMDB、MassBank、PubChem数据库比对,活性组分Ⅲ在正、负离子模式下共鉴定31个化合物,主要包括胺类、酰胺类、有机酸、生物碱、苯甲酸盐等化合物;而活性组分Ⅵ在正、负离子模式下鉴定34个化合物,主要包括有机酸、酰胺类、酚类、生物碱、苯甲酸盐等化合物。本试验结果为研制高效、安全和廉价的防治奶牛胎衣不下的中药制剂奠定基础。 To identify the chemical constituents of active componentsⅢandⅥof semen plantaginis by stir-frying with wine on treatment of retained fetal membranes in cows by LC-MS/MS.Chromatograpic separation was performed,Hyper gold C18 column(100.0 mm×2.1 mm,1.9μm)was used as chromatographic column,acetonitrile(containing 0.1%formic acid)-water(containing 0.1%formic acid,5%acetonitrile)was used as mobile phase for gradient elution,the flow rate was 0.3 mL/min,the column temperature was 40℃.LC-MS/MS was developed for rapid and high-throughput screening of the preliminary chemical profile of active componentsⅢ,Ⅵin positiveion and negativeion modes.The according to the accurate charge-mass ratio and the MS/MS data,Metlin,HMDB,MassBank and PubChem databases,the fragmentation regularities of mass spectra.In positiveion and negativeion modes,the thirty-one chemical components were found in active componentsⅢ,mainly including amides,organoheterocyclic compound,benzenoids,organic acid,alkaloid and amine.The thirty-four chemical components were found in active componentsⅥ,mainly including organic acid,amides,organoheterocyclic compound,benzenoids,amine and phenols.The results of this study laid a practical foundation for the development of high efficiency,safe and cheap Chinese medicine preparation for preventing and controlling the retained fetal membranes in the dairy cows.
