摘要
目的建立HPLC外标法快速测定人血清中伏立康唑浓度。方法人血清经乙腈沉淀后,以乙腈-水-冰醋酸-三乙胺(45∶55∶0.6∶0.4)为流动相,流速为1.4mL·min^(-1),色谱柱为Ultimate AQ-C_(18)(250mm×4.6mm,5μm),柱温为35℃,紫外检测波长为254nm。结果血清内源性杂质不干扰伏立康唑测定,伏立康唑的线性范围为0.5~8.0μg·mL^(-1),日内、日间精密度(RSD)均小于5%,血样稳定性良好。结论该方法操作简便、快速、准确,可用于伏立康唑的血清药物浓度监测。
OBJECTIVE To establish a rapid HPLC external standard method for determination of voriconazole in human serum.METHODS Human serum was treated by acetonitrile.Separation was carried out on the column of Ultimate AQ-C 18(250mm×4.6mm,5μm)at 35℃,with the mobile phase(acetonitrile-water-acetic acid-triethylamine,45∶55∶0.6∶0.4)at a flow rate of 1.4mL·min-1.The detecting wavelength was 254nm.RESULTS The intrinsic impurity of serum did not interfere with the determination of analytes.The liner range of voriconazole was 0.5-8.0μg·mL-1.The precision of inter day and intra day were less than 5%.The stability of unprocessed and processed samples was good.CONCLUSION The established HPLC method is simple,fast,accurate and can be applied to the TDM of voriconazole.
