基于RNA-Seq技术的乒乓菊转录组分析 被引量:1

Transcriptome Analysis of Dendranthema morifolium cv.'pompon'Based on RNA-Seq Technology

摘要 为开展乒乓菊基因功能分析、表型差异、分子标记开发和遗传多样性等研究,本试验以‘绿乒乓’和‘粉乒乓’的花朵和叶片混合样品为试验材料,通过转录组测序分析,经de novo组装后获得133200条Unigenes,进一步利用6大公共数据库对其进行注释,共注释64601条Unigenes,并基于转录组数据开展SSR和SNP位点预测。结果表明:有25462条Unigenes参与了KEGG代谢通路,包括与花色、花期和药用成分代谢等相关的途径;共预测到1444个转录因子,分属于35个家族,包括在植物的生长发育、生物合成、抗非生物和生物胁迫等方面发挥重要作用的AP2/ERF、C2C2、bHLH、WRKY等转录因子家族,以及LBD、MADS和TCP等与花色素苷代谢、花和根系发育、植物进化和发育密切相关的转录因子家族。此外,从13014条Unigenes序列中搜索到14905个SSR位点;并分别从‘绿乒乓’和‘粉乒乓’中挖掘到1081925个和1077818个SNP位点。本研究可为乒乓菊功能基因挖掘和分子标记开发提供基础。 In order to carry out research on functional analysis,phenotypic difference,molecular marker development and genetic diversity of pompon.In this experiment,the mixed samples of flowers and leaves of’Green pompon’and’Pow pompon’were used as experimental materials.After transcriptome sequencing analysis,133200 Unigenes were obtained after de novo assembly,and further commented by six public databases.A total of 64601 Unigenes were annotated,SSR and SNP locus predictions were performed based on transcriptome data.The results showed that there were 25462 Unigenes involved in the KEGG metabolic pathway,including pathways related to flower color,flowering and medicinal component metabolism.A total of 1444 transcription factors were predicted,belonging to 35 families,including AP2/ERF,C2C2,bHLH and WRKY,which play an important role in plant growth and development,biosynthesis,antibiotic and biotic stress.As well as LBD,MADS and TCP,which are closely related to anthocyanin metabolism,flower and root development,plant evolution and development.In addition,14905 SSR loci were searched from 13014 Unigenes sequences,1081925 and 1077818 SNP loci were mined from’Green pompon’and’Pow pompon’respectively.This study can provide a basis for functional gene mining and molecular marker development in Dendranthema morifolium cv.’pompon’.
