基于SLAF-Seq技术的广金钱草SNP位点开发及遗传分析

SNP Loci Development and Genetic Analysis of Desmodium styracifolium(Osbeck)Merr Based on SLAF-Seq Technology

为分析不同来源广金钱草的遗传关系,本研究利用SLAF-seq技术对来自广东、广西、海南的20份广金钱草样品进行测序,共获得SLAF标签322360个,鉴定到72395个多态性SLAF标签。进行过滤后开发得到SNP多态标记220704个,分析得到94505个高一致性的群体SNP。通过对20份不同来源的广金钱草个体完成系统进化树、群体结构分析,结果表明,20份广金钱草来源于同一祖先,但由于地理隔离的影响使得个体间发生了遗传分化。通过系统进化树分析发现分布在广东、广西及海南琼中、屯昌的广金钱草具有较近的亲缘关系。研究结果为种质保护和遗传改良提供参考。

To analyze the genetic relationship among different cultivars,20 sites of Desmodium styracifolium(Osbeck)Merr.distributed in Guangdong,Guangxi and Hainan Province were used as experiment materials for sequencing by specific-locus amplified fragment sequencing(SLAF-seq).In total,we obtained 322360 SLAF tags,including 72395 polymorphic SLAF tags.By screening from all SNP,obtained 94505 high consistency SNP sites were obtained based on the analysis of total 220704 SNP.Phylogenetic relationship and population structure analysis of20 individuals of D.styracifolium were analyzed using mathematical statistical method based on screening SNP.The results showed that these D.styracifolium in 20 different sites all originate from the same ancestor,but due to this factor that geographical location makes the D.styracifolium produced genetic differentiation.Phylogenetic tree analysis showed that,D.styracifolium growing in Guangdong,Guangxi,Hainan Qiongzhong and Hainan Tunchang have a close relationship.The results will provide reference for the protection and genetic improvement of D.styracifolium’s germplasm.
