菜豆DUS测试标准品种SSR遗传多样性分析 被引量:5

摘要 为了对《植物新品种特异性,一致性和稳定性测试指南菜豆》标准品种选用的合理性进行评价,为测试指南进一步的修订和完善提供分子层面的参考依据,本研究对菜豆DUS测试标准品种的遗传多样性进行分析。结果显示,33对核心引物在26个标准品种中共检测到214个等位变异,平均每对引物检测到了6.49个等位变异,该套引物的Shannon's信息指数在0.5158~2.1257之间,均值是1.2957;多态信息含量(PIC)处于0.2480~0.8609之间,平均值为0.6040。26个标准品种的遗传相似系数在0.03~0.85之间,平均值为0.31,表明品种间遗传多样性较丰富,标准品种选择较为合理。其中仅‘双阳大花皮’和‘大家花雀蛋间’的遗传相似系数大于0.8,且二者在测试指南中均只指示一个性状,可以根据这一结果开展进一步田间对比试验,尝试减少标准品种数量,提高DUS测试效率。 In order to evaluate the rationality of the selection of the sample varieties in French bean DUS testing,and provide a molecular basis for revision and improvement the test guide,this study analyzed the genetic diversity of the sample varieties based on the SSR fingerprint database.The results showed that 33 pairs of core primers detected 214 alleles in 26 varieties,and an average of 6.49 alleles were detected for each primer.The Shannon’s information index of those primers ranged from 0.5158 to 2.1257,with an average of 1.2957.The polymorphic information content(PIC)ranged from 0.2480 to 0.8609 with an average of 0.6040.The genetic similarity coefficients of the 26 sample varieties ranged from 0.03 to 0.85,with an average of 0.31,indicating that the genetic diversity among the varieties was rich,and the selection of sample varieties was more reasonable.Among them,only the genetic similarity coefficient between’Shuangyangdahuapi’and’Dajiahuaquedan’is 0.8,and both indicate only one trait in the standard.According to this result,field comparison test can be carried out to reduce the number of standard varieties and reduce the workload of DUS testing to a certain extent.
