离子色谱法测定岩藻聚糖中L-岩藻糖 被引量:5

Determination of L-fucose in fucoidan by ion chromatography

摘要 建立测定岩藻聚糖中L-岩藻糖的离子色谱方法。对包括色谱柱、流动相等色谱条件进行了优化,选用DIONEX CarboPac PA10型(250 mm×4 mm)色谱柱,以H2O-250 mmol/L NaOH溶液(90∶10)为流动相,流量为1.00 mL/min,柱温为30℃,用脉冲安培检测器检测。在该条件下,L-岩藻糖的质量浓度在0.11~44.80μg/mL范围与色谱峰面积线性良好,相关系数为0.9924,检出限为0.05μg/mL,定量限为0.10μg/mL。样品加标平均回收率为91.79%,相对标准偏差为2.77%(n=6)。该法检测岩藻聚糖中L-岩藻糖,操作简单、快速,灵敏度高,测定结果准确。 An ion chromatographic method was developed for the determination of L-fucose in fucoidan.After the optimization of chromatographic conditions such as column,mobile phase and so on were optimized,the samples were operated on DIONEX CarboPac PA10 Column(250 mm×4 mm)by using a mixture of H2O and 250 mmol/L sodium hydroxide solution(90∶10)as the mobile phase with the flow rate of 1.00 mL/min.The column temperature was 30℃and the detector was pulse ampere detector.A good linearity was observed in the concentration range of 0.11-44.80μg/mL with the correlation coefficient of 0.9924.The detection limit and the quantitative limit were 0.05μg/mL and 0.10μg/mL,respectively.The average recovery of L-fucose was 91.79%with the relative standard deviation of 2.77%(n=6).The results indicated that ion chromatographic method was a simple,fast,sensitive,accurate and effective method for the detection of L-fucose in fucoidan.
