褪黑素对湖羊精液低温保存效果的影响 被引量:3

Effect of melatonin on low temperature preservation of Hu Sheep semen

摘要 为研究褪黑素对湖羊精液4℃保存效果的影响,在前期筛选的湖羊精液稀释液中添加不同浓度(0.00、0.02、0.04、0.06、0.08 g/L)的褪黑素进行4℃冷藏试验,检测湖羊精子活力和顶体完整率,以及精液中过氧化氢酶(CAT)活力和丙二醛(MDA)含量。结果:与对照组相比,添加0.02 g/L褪黑素组在保存0、24、48、72、96 h的精液中MDA含量极显著降低(P<0.01);0、24、48、72 h精液中CAT酶活力极显著提高(P<0.01);48 h精子活力和顶体完整率极显著提高(P<0.01)。综合比较表明,在湖羊精液稀释液中添加0.02 g/L褪黑素,在48 h内能够显著提高湖羊精液4℃稀释保存的品质。 In order to determine the effect of melatonin on preservation of Hu Sheep semen stored at 4℃,different concentrations of melatonin(0.00/control,0.02/Ⅰ,0.04/Ⅱ,0.06/Ⅲ,0.08/Ⅳg/L)were added to the pre-screened semen diluent for Hu Sheep semen,and the sperm motility,acrosome integrity rate,catalase(CAT)activity and malondialdehyde(MDA)content were tested of the semen preserved at 4℃.The results showed that,compared with the control group,the MDA content in the semen of GroupⅠwas significantly reduced at 0,24,48,72,96 h(P<0.01)of preservation;the CAT activity was significantly enhanced at 0,24,48,72 h(P<0.01);and the sperm motility and acrosome integrity rate were significantly raised at 48 h(P<0.01).It could concluded that addition of melatonin at the concentration of 0.02 g/L in the dilution improve the preservation effect of Hu Sheep semen at 4℃for 48 hours.
