固定时间的动员方案与外周血造血干细胞分布规律分析 被引量:4

摘要 目的通过观察固定时间的动员方案供者外周血干细胞的分布规律,进一步探讨干细胞最佳的采集时机。方法采用固定的造血干细胞动员方案,即供者皮下注射重组人粒细胞集落刺激因子10μg/(kg·d),每12 h一次(8:00、20:00各一次)。采集供者动员后第4天多个时间点的外周血,用于测定血常规及CD34+细胞,绘图并观察分布规律。结果采用固定时间的造血干细胞动员方案,干细胞均在12:00达到峰值,供者动员后第5天10:00至15:00为最佳采集时机。结论采用固定时间的动员方案能预期获得干细胞在外周血分布的规律以及采集的最佳时机,建立干细胞采集的临床路径。 Objective To find out the distribution regularities of peripheral blood stem cells by using the steady-time mobilisation scheme,and to explore the optimal timing of the collection of the hematopoietic stem cells.Methods The steady-time hematopoietic stem cell mobilization scheme was adopted.The donors were subcutaneously injected with the recombinant human granulocyte colony stimulating factor 10μg/(kg·d),every 12 h(8:00,20:00 every day).Peripheral blood of the donors which was collected at multiple time points on the 4th day after the mobilization was used to check the blood routine examination and CD34+cells.Then we made charts to observe the distribution regularities.Results Stem cells all peaked at 12:00 by adopting the steady-time mobilisation scheme,so 10:00 to 15:00 on the 5th day after donor mobilization was the optimal timing of collection.Conclusion the distribution regularities of peripheral blood stem cells and the optimal timing of collection can be obtained by adopting the steady-time mobilisation scheme,so that the clinical pathway of the hematopoietic stem cell collection can be established.
