摘要
为筛选优化褐黄木耳母种培养基配方,以菌丝日均长速为测量指标,菌丝长势为参考项,通过单因素试验,筛选出母种培养基最佳碳源和最佳氮源,并运用正交试验法优化配方。结果表明,褐黄木耳母种培养基最佳配方为蔗糖2%,酵母浸膏0.5%,KH2PO40.3%,MgSO4·7H2O 0.15%,琼脂2%,pH自然。利用该配方培养,褐黄木耳菌丝色白、密实,平均生长速度3.82 mm/d,高于正交试验和单因素试验结果。
Taking the average daily growth rate of hypha as the measurement index and hypha growth potential as the reference item,the best carbon source and the best nitrogen source of Auricularia fuslosuccinea stock culture medium were selected through single factor experiment,and the formula of stock culture medium was optimized by orthogonal test.The results showed that the optimal formula of stock culture medium for Auricularia fuslosuccinea was:sucrose 2%,yeast extract 0.5%,KH2PO40.3%,MgSO4·7H2O 0.15%,agar 2%,and natural pH.
