大孔树脂纯化油菜叶多糖的工艺及其抗氧化活性研究 被引量:6

摘要 研究大孔树脂对油菜叶多糖的纯化工艺,并就纯化后的多糖对DPPH、ABTS、羟基自由基的清除作用进行考察。结果表明:AB-8型大孔树脂为最佳纯化树脂;最佳吸附条件为pH4.0、上样浓度6.0 mg/mL、上样速率1.0 BV/h;最佳解吸条件为乙醇体积分数70%、洗脱剂用量3.0BV、洗脱速率2.0BV/h。纯化后的油菜叶多糖含量从9.84%提高到59.56%,对几种自由基都有很好清除作用,且随着多糖浓度增大而增强。 The purification process of polysaccharides from leaves of Brassica napus L.was studied,and the scavenging effects of the purified polysaccharides on DPPH,ABTS and hydroxyl radicals were investigated.The results showed that AB-8 was the best resin for the purification of polysaccharides.The best dynamic adsorption conditions were:pH 4.0,the concentration of sample 6.0 mg/mL,the sample loading speed 1.0 BV/h.The best dynamic desorption conditions were:the ethanol concentration 70%,the amount of eluent 3.0 BV,the flow rate 2.0 BV/h.The content of the purified polysaccharide increased from 9.84%to 59.56%,which had a good scavenging effect on several free radicals and increased with the increase of the increase of the concentration of polysaccharide.
