摘要 目的研究参菊洗剂的抗炎作用机制。方法采用二甲苯致小鼠耳廓肿胀、醋酸致小鼠腹腔毛细血管通透性增加、角叉菜胶致大鼠足跖肿胀3种炎症模型,以0.1 g/mL洁尔阴洗液为对照,评价不同浓度(0.054、0.108、0.216 g/mL)参菊洗剂的抗炎作用,比较各组血清和足跖组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、前列腺素E2(PGE2)、肿瘤坏死因子α(TNF-α)指标的水平。结果与模型对照组比较,参菊洗剂各剂量和阳性对照组小鼠肿胀抑制率和腹腔毛细血管通透性抑制率均升高,参菊洗剂各剂量和阳性对照组大鼠致炎后1、2、3、4 h大鼠足跖肿胀度均降低,血清中SOD水平升高,MDA和TNF-α水平均降低,足跖组织中SOD水平升高,MDA、PGE2和TNF-α水平均降低,差异有统计学意义(P<0.01~0.05)。与阳性对照组比较,参菊洗剂高剂量组小鼠肿胀抑制率和腹腔毛细血管通透性抑制率升高,参菊洗剂高剂量组大鼠在致炎后1、2、3、4 h足跖肿胀度降低,血清和足跖组织中MDA、TNF-α水平均降低;参菊洗剂中、高剂量组大鼠血清和足跖组织中SOD水平升高,差异有统计学意义(P<0.05)。结论参菊洗剂具有抗炎作用,其抗炎机制与提高SOD活性,抑制脂质过氧化和增强清除自由基、抑制MDA、PGE2、TNF-α炎症因子的合成与释放有关。 Objective To study the anti-inflammatory mechanism of Shenju lotion.Methods Three kinds of inflammation models were used:auricle swelling induced by xylene in mice,peritoneal capillary permeability increased by acetic acid in mice,and paw swelling induced by carrageenan in rats.0.1 g/ml Jieeryin lotion was used as control,and different concentrations(0.054,0.108,0.216)of the lotion were evaluated.The levels of superoxide dismutase(SOD),malondialdehyde(MDA),prostaglandin E2(PGE2)and tumor necrosis factor-α(TNF-α)in serum and plantar tissue of inflammatory rats were measured.Results Compared with the mice model group,the swelling inhibition rate and peritoneal capillary permeability inhibition rate of each dose of Shenju lotion and the positive control group were all increased.After 1,2,3 and 4 hr(s)after the inflammation of rats,in each dose of Shenju lotion and the positive control group,the swelling degree of rat paw was decreased,the level of SOD in the serum was increased,the level of MDA and TNF-αin the serum was decreased;the level of SOD in the plantar tissue was increased,the levels of MDA,PGE2 and TNF-αin the plantar tissue were decreased,and the differences were statistically significant(P<0.01,0.05).Compared with the positive control group,the swelling inhibition rate and peritoneal capillary permeability inhibition rate of mice in the high-dose group of Shenju lotion were increased;and the rats in the high-dose Shenju lotion group,at 1,2,3 and 4 hr(s)after the inflammation,the swelling degree of plantar was decreased,the levels of MDA and TNF-αin the serum and plantar tissue were decreased;SOD levels in the serum and plantar tissue of rats in the mid-and high-dose Shenju lotion groups were increased,and the difference was statistically significant(P<0.05).Conclusion Shenju lotion has anti-inflammatory effect.Its anti-inflammatory mechanism is related to improving SOD activity,inhibiting lipid peroxidation,enhancing scavenging free radicals,and inhibiting synthesis and release of the inflammatory factors MDA,PGE2 and TNF-α.
