摘要
目的探讨浙江献血者中丙型肝炎病毒(HCV)感染与KIR基因的相关性。方法选择2011年10月-2017年8月在浙江省血液中心的献血者,经血液核酸筛查HCV RNA阳性的样本,以健康人群为对照样本。HCV RNA阳性样本和健康对照样本均用PCR-SSO的方法获得KIR基因频率,根据基因分布情况进行KIR基因型判断。用qPCR的方法分析KIR2DL1和KIR2DL3基因拷贝数差异。结果HCV感染者KIR基因和基因型频率与健康对照者分布差异无统计学意义(P>0.05),而KIR2DL3拷贝数在健康对照者较HCV感染者显著升高,差异有统计学意义(P<0.01)。结论在浙江献血者中尚未发现HCV感染与KIR基因和基因型分布有相关性,而KIR2DL3基因在健康人群中可能通过基因数量对HCV的感染有一定的影响。
Objective To find out the association of KIR genes with Hepatitis C Virus infection in Zhejiang blood donors.Methods Blood donors who donated blood from October 2011 to August 2017 were selected for HCV RNA-positive samples by nucleic acid amplification technology in Blood Center of Zhejiang Province.Healthy subjects were selected as population control.KIR gene frequency was obtained by PCR-SSO in both HCV RNA positive samples and control samples,and KIR genotype was determined according to gene distribution.The gene copy number differences of KIR2DL1 and KIR2DL3 were analyzed by qPCR.Results There were no significant differences in the distribution of KIR gene and genotype frequency between HCV-infected patients and the controls(P>0.05),while the copy number of KIR2DL3 in the controls was significantly higher than that in HCV-infected patients,with the difference statistically significant(P<0.01).Conclusion HCV infection has not been found to be associated with KIR gene distribution in Zhejiang blood donors,and KIR2DL3 gene may have a certain effect on viral infection through gene number in healthy people.
