摘要
不同品种及外植体对不定芽诱导和植株再生存在一定的差异,不同的培养条件对猕猴桃组培苗不定芽的再生也会产生一定的影响。本文以‘米良1号’、‘红阳’、‘金艳’猕猴桃为试验材料,研究外植体类型、品种和培养基等对组培苗不定芽再生的影响。结果表明,3个猕猴桃品种的叶片和叶柄均可诱导出愈伤组织,并再分化出不定芽;茎尖和茎段则不经过愈伤组织阶段,直接分化出不定芽。品种和培养基对叶片、叶柄愈伤组织诱导率的影响差异显著,对叶片、叶柄不定芽诱导率的影响差异不显著;培养基对茎尖、茎段不定芽诱导率差异均显著,但品种间差异不显著。
Different varieties and explants had different effects on adventitious bud induction and plant regeneration.The regeneration of adventitious shoots in tissue culture of kiwi fruit was also affected by different culture conditions.In this study,kiwifruit cultivars of‘Miling-1’,‘Hongyang’and‘Jinyan’were used as materials to study the effect of explant types,varieties and culture conditions on adventitious bud regeneration.The results showed that callus tissue was induced from leaves and petioles and then adventitious buds could be differentiated from callus tissue for three varieties.The adventitious buds could be directly induced from shoot tips and stems without a callus stage.There were significant differences in the callus induction rates of leaves and petioles among varieties and culture mediums,but no significant difference in induction rates of adventitious buds was found.There were significant differences in the induction rates of adventitious buds in shoot tips and stems among culture mediums,but no significant difference among varieties was found.
