摘要 目的:探究超声-微泡介导的miR-126通过调节CRK对乳腺癌细胞活力的影响。方法:qPCR检测miR-126在乳腺癌组织和乳腺癌细胞系中的表达,并分析其表达与乳腺癌临床特征的相关性。利用超声技术制备脂质微泡以探究超声-微泡介导的miR-126对乳腺癌细胞增殖、侵袭和凋亡的影响。EdU染色、Transwell和流式细胞术分别用于检测MDA-MB-231细胞增殖、侵袭和凋亡情况。qPCR和Western blot检测CRK的mRNA和蛋白水平。此外,Western blot检测凋亡相关蛋白Bcl-2、Bax、c-Caspase-3和pro-Caspase-3的蛋白水平。结果:miR-126在乳腺癌组织和细胞中表达下调。过表达miR-126抑制乳腺癌细胞活力和侵袭能力,促进细胞凋亡。超声-微泡介导的miR-126进一步增强了miR-126对乳腺癌细胞活力的抑制作用。另外,miR-126通过调节CRK从而抑制乳腺癌细胞的活力。结论:miR-126过表达可抑制乳腺癌细胞的活力,超声-微泡介导的miR-126进一步增强了miR-126对乳腺癌细胞活力的抑制作用。本研究为乳腺癌的治疗提供了新的分子靶标和治疗途径。 Objective:To explore the effect of ultrasound-microbubble-mediated miR-126 on the activity of breast cancer cells by regulating CRK.Methods:The expression of miR-126 in breast cancer tissues and breast cancer cell lines were detected by qPCR,and its correlation with breast cancer clinical features was analyzed.Ultrasound technology was used to extract lipid microbubbles to explore the effect of ultrasound-microbubble-mediated miR-126 on breast cancer cell proliferation,invasion and apoptosis.EdU staining,Transwell and flow cytometry were used to detect cell viability,invasion and apoptosis,respectively.qPCR and Western blot were used to detect the expression levels of CRK mRNA and protein.And apoptosis-related proteins Bcl-2,Bax,c-Caspase-3 and pro-Caspase-3 were measured using Western blot.Results:The expression of miR-126 was down-regulated in breast cancer tissues and cells.Overexpression of miR-126 inhibited the viability and invasion ability of breast cancer cells and promoted apoptosis.Ultrasound-microbubble-mediated miR-126 further enhanced the inhibitory effect of miR-126 on breast cancer cell activity.In addition,miR-126 regulated CRK to inhibit the activity of breast cancer cells.Conclusion:Overexpression of miR-126 can inhibit the activity of breast cancer cells.Ultrasound-microbubble mediated miR-126 further enhances the inhibitory effect of miR-126 on breast cancer cell activity.This study provides novel molecular targets and therapeutic approaches for the treatment of breast cancer.
