摘要 目的:探讨蛋氨酸脑啡肽(MENK)联合紫杉醇(PTX)对肺癌细胞A549的增殖和凋亡影响。方法:CCK-8法检测各实验组对细胞增殖的抑制作用;利用中效原理对药物联合应用的效应进行评价;Hoechst33258荧光染色实验观察细胞凋亡形态;流式细胞术检测各实验组的细胞凋亡率。结果:MENK和PTX均能抑制A549细胞的增殖,并且MENK联合PTX组抑制率高于单独用药组;在整个效应范围中,MENK与PTX的药物合用指数(CI)<1,两药联合应用效应为协同作用;Hoechst33258荧光实验发现,MENK与PTX组均出现凋亡形态学变化,MENK联合PTX组细胞凋亡数量最多;流式细胞术检测空白对照组、MENK组、PTX组、MENK联合PTX组的细胞凋亡率分别是1.53%、6.23%、6.04%、13.22%,联合用药组的细胞凋亡率明显高于单独用药组。结论:MENK与PTX均能抑制肺癌细胞A549的增殖,并诱导其凋亡,两药联合应用具有协同作用,显著增强抑制细胞增殖和诱导细胞凋亡的能力。 Objective:To investigate the effects of methionine enkephalin(MENK)combined with paclitaxel(PTX)on proliferation and apoptosis of lung cancer cell A549.Methods:The inhibitory effect of each experimental group on cell proliferation was detected by CCK-8 method.The effect of drug combination was evaluated by the median-effect principle;the apoptosis morphology was observed by Hoechst33258 fluorescence staining.The apoptosis rate of each experimental group were detected by flow cytometry.Results:MENK and PTX could inhibit the proliferation of A549 cells,and the inhibition rate of MENK combined with PTX group was higher than that of the drug alone group.In the whole range of effects,the drug combination index(CI)of MENK and PTX was<1,and the combined effect of the two drugs was synergistic.Hoechst33258 fluorescence assay showed that apoptotic morphological changes were observed in both MENK and PTX groups,and the number of apoptosis was the highest in MENK combined with PTX group.Flow cytometry showed that the apoptosis rates of the blank control group,MENK group,PTX group,MENK combined with PTX group were 1.53%,6.23%,6.04%and 13.22%,respectively.The apoptosis rate of the combination group was significantly higher than that of the single medication group.Conclusion:Both MENK and PTX can inhibit the proliferation of lung cancer cell A549 and induce its apoptosis.The combination of the two drugs has synergistic effect and significantly enhances the ability of inhibiting cell proliferation and inducing apoptosis.
