咳喘宁对RSV诱导哮喘大鼠的治疗作用及对IL-33表达的影响 被引量:4

摘要 目的:探讨咳喘宁对病毒诱发哮喘大鼠的治疗作用及对IL-33表达的影响。方法:以呼吸道合胞病毒(RSV)滴鼻诱导+卵清白蛋白致敏的方法制作哮喘模型,实验动物随机分为正常组、模型组、咳喘宁高剂量组、咳喘宁中剂量组和咳喘宁低剂量组,各组分别给予生理盐水和相应药物处理。末次给药后进行大鼠气道高反应性测定;HE染色和Masson染色分别观察肺组织病理学改变和气道下胶原沉积情况;ELISA法检测血清中IL-33水平,qPCR法检测肺组织IL-33 mRNA表达。结果:与正常组相比,模型组大鼠哮喘症状严重,气道反应性升高,肺组织结构破坏,气道下胶原沉积增多(P<0.05),血清中IL-33蛋白和肺组织IL-33 mRNA表达显著升高(P<0.05);与模型组相比,咳喘宁高、中、低剂量组哮喘症状明显减轻(P<0.05),气道反应性降低(P<0.05),肺组织病理改变轻微,气道下胶原沉积减少(P<0.05),血清中IL-33蛋白和肺组织IL-33 mRNA表达明显下调(P<0.05),其中以咳喘宁高剂量组的改变最为显著(P<0.05)。结论:咳喘宁对RSV诱导哮喘大鼠的治疗作用显著,可有效减轻气道炎症,抑制气道重塑,阻断哮喘病理改变,其机制可能与降低血清与肺组织中IL-33表达有关。 Objective:To investigate therapeutic effect of Kechuanning on virus-induced asthmatic rats and its effect on IL-33 expression.Methods:Asthma model was established by method of respiratory syncytial virus(RSV)nasal induction and ovalbumin sensitization,experimental animals were randomly divided into normal group,model group,Kechuanning high dose group,Kechuanning middle dose group and Kechuanning low-dose group,each group was given control saline and corresponding drugs.Airway hyperresponsiveness was measured after the last administration;pathological changes of lung tissue and collagen deposition of airway of lung tissue were observed by HE staining and Masson staining;level of IL-33 in serum was detected by ELISA,and expression of IL-33 mRNA in lung tissue was detected by qPCR.Results:Compared with normal group,model group had severe asthma symptoms and increased airway responsiveness,with lung tissue structure damaged and increased collagen deposition under the airway(P<0.05);expression of IL-33 protein in serum and IL-33 mRNA in lung tissue were significantly increased(P<0.05);compared with model group,symptoms of asthma in the high,middle and low dose groups of Kechuanning were significantly alleviated(P<0.05),airway responsiveness was decreased(P<0.05),staining of lung tissue ws mild,and collagen deposition decreased under airway(P<0.05);expressions of IL-33 protein in serum and IL-33 in lung tissue were mRNA were significantly down-regulated(P<0.05).Among them,change of Kechuanning high dose group was the most significant(P<0.05).Conclusion:Kechuanning has a significant therapeutic effect on RSV-induced asthmatic rats,which can effectively alleviate airway immune allergic inflammation,inhibit airway remodeling and block pathological changes of asthma,the mechanism may be related to decrease of IL-33 expression in serum and lung tissue.
