摘要
为进一步确定湖北恩施地区茶园蓟马的种类,本文利用DNA条形码技术,对该地区茶叶上的蓟马进行鉴定,通过COI基因的通用引物,扩增mtDNA COI基因并测序、比对和聚类分析。结果表明,该茶园中的蓟马与茶棍蓟马mtDNA COI(NC_037839.1)相似度达100%,为茶棍蓟马,系统发育树表明茶棍蓟马的mtDNA COI基因与豆喙蓟马在同一个进化枝上,其bootstrap值为75%。
In order to further determine the species of thrips in the tea garden of Enshi,Hubei Province,DNA barcoding technology has been used to identify thrips on tea in this area.By using the universal primers,the mtDNA COI gene was amplified and sequenced,aligned and cluster analyzed.The results showed that the similarity of the mtDNA COI between the thrips in Hefeng tea garden and Dendrothrips minowai(NC_037839.1)was 100%.Phylogenetic analysis showed that the mtDNA COI gene of Dendrothrips minowai was on the same clade where Mycterothrips glycines locates,with the bootstrap value of 75%.
