女贞子的不同炮制品配伍对二至丸中12种成分含量的影响 被引量:5

Effects of Compatibility of Different Prepared Products of Ligustrum lucidum on the Contents of Twelve Components in Erzhi Pills

摘要 目的:建立HPLC同时测定二至丸中12种成分含量的方法,探讨女贞子的不同炮制品配伍对二至丸成分含量的影响。方法:色谱柱:Kromasil 100-5 C18柱(250 mm×4.6 mm,5μm);流动相:乙腈-0.2%磷酸溶液,梯度洗脱;流速:1.0 mL/min;柱温:30℃;检测波长:225、340 nm。测定女贞子不同炮制品配伍的二至丸中12种成分的含量。结果:羟基酪醇、红景天苷、女贞酸、酪醇、绿原酸、松果菊苷、毛蕊花糖苷、异去甲蟛蜞菊内酯、蟛蜞菊内酯、特女贞苷、女贞苷G13、oleonuezhenide在相应的进样量范围内线性关系良好(r≥0.9995),平均加样回收率为93.9%~103.4%,RSD均<2.50%。与二至丸Ⅰ(生女贞子、墨旱莲)比较,二至丸Ⅱ(酒蒸女贞子、墨旱莲)和二至丸Ⅲ(清蒸女贞子、墨旱莲)中女贞酸的含量明显增加,毛蕊花糖苷含量明显降低;羟基酪醇、酪醇、绿原酸、松果菊苷、异去甲蟛蜞菊内酯、蟛蜞菊内酯含量无明显变化。结论:该方法操作简便、准确、稳定、重复性好,可用于二至丸的质量控制。炮制对二至丸中环烯醚萜苷和苯乙醇类成分含量具有明显影响。 Objective:To establish a method for simultaneous quantitative determination of 12 components in Erzhi pills by HPLC,to study the effect of compatibility of different prepared products of Ligustrum lucidum on the contents of 12 components in Erzhi pills.Methods:The chromatographic column was Kromasil 100-5 C18(250 mm×4.6 mm,5μm)column;Mobile phase was acetonitrile(A)-0.2%phosphoric acid aqueous solution(B)with gradient elution.The flow rate was 1.0 mL/min,column temperature was 30℃and detection wavelength was 225,340 nm.The contents of 12 components in Erzhi pills compatible with different processed products of Ligustrum lucidum were determined.Results:Hydroxytyrosol,salidroside,nuezhenidic acid,tyrosol,chlorogenic acid,echinacoside,verbascoside,isodemethylwedelolactone,demethylwedelolactone,specnuezhenide,nuezhenoside G13,oleonuezhenide had good linear relationships within the corresponding injection volume range(r≥0.9995),the average recovery rate of the sample was 93.9%~103.4%,RSD was less than 2.50%.Compared with Erzhi pillsⅠ(raw Ligustrum lucidum and Eclipta prostrata),the content of nuezhenidic acid in Erzhi pillsⅡ(wine steamed Ligustrum lucidum and Eclipta prostrata)and Erzhi pillsⅢ(steamed Ligustrum lucidum and Eclipta prostrata)was significantly increased and the content of verbascoside decreased obviously,while the contents of hydroxytyrosol,tyrosol,chlorogenic acid,echinacoside,isodemethylwedelolactone and demethylwedelolactone were no significant changes.Conclusion:The method is simple,accurate,stable and reproducible,can be used for quality control of Erzhi pills.Processing has obvious influence on the contents of iridoid glycoside and phenylethanol in Erzhi pills.
