摘要
【目的】探讨加味苓桂术甘汤改善代谢综合征(MS)大鼠胰岛素抵抗的作用和机制。【方法】从50只健康雄性SD大鼠中随机选取10只作为空白组,其余40只采用高脂高糖高盐饲料喂养方法建立代谢综合征大鼠模型。将符合代谢综合征标准的35只大鼠随机分为模型组(11只)、中药组(12只)、西药组(12只),继续给予高脂高糖高盐饲料喂养,并分别对应给予生理盐水、加味苓桂术甘汤、盐酸罗格列酮混悬液灌胃8周。造模与给药期间,空白组均给予普通饲料喂养。检测大鼠体质量、血压[收缩压(SBP)、舒张压(DBP)],血脂[总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)],空腹血糖(FBG),空腹胰岛素(FINS),脂肪组织游离脂肪酸(FFA)、瘦素(LP)、脂联素(ADP)、内脂素(VF)、过氧化物酶体增殖剂激活受体γ(PPAR-γ),骨骼肌组织葡萄糖转运蛋白4(GLUT-4)水平,计算胰岛素抵抗指数(IRI)和Lee’s指数。【结果】与空白组比较,模型组大鼠的体质量、SBP、DBP、FBG、FINS、TC、TG、LDL-C、FFA、LP、VF、IRI、Lee’s指数水平明显升高,HDL-C、ADP、GLUT-4、PPAR-γ水平明显降低(均P <0.05);与模型组比较,中药组和西药组大鼠体质量、SBP、DBP、FBG、FINS、TC、TG、LDL-C、FFA、LP、VF、IRI、Lee’s指数水平明显降低,HDL-C、ADP、GLUT-4、PPAR-γ水平明显升高(均P <0.05),且2个治疗组比较,差异均无统计学意义(P> 0.05)。【结论】加味苓桂术甘汤可改善代谢综合征大鼠的糖脂代谢紊乱和胰岛素抵抗,其机制可能与调节内脏脂肪组织FFA、LP、ADP、VF、PPAR-γ等脂肪细胞因子和骨骼肌GLUT-4表达有关。
Objective To explore the effects and mechanism of Modified Linggui Zhugan Decoction in improving insulin resistance in rats with metabolic syndrome. Methods Ten rats were randomly selected from 50 healthy male Sprague-Dawley rats as blank group,the other 40 rats were fed with high-fat,high-sugar and high-salt fodder to establish MS model. And then 35 rats met the MS standard were randomly divided into model group(11 rats),Chinese medicine group(12 rats)and western medicine group(12 rats),and were all given continue high-fat,high-sugar and high-salt fodder and respectively corresponding given normal saline,modified Linggui Zhugan Decoction,rosiglitazone for 8 weeks. During the modeling and medication,the blank group was given ordinary fodder feeding. The body mass,blood pressure [systolic blood pressure(SBP),diastolic blood pressure(DBP)],blood lipids [total cholesterol(TC), triglycerides(TG), high-density lipoprotein cholesterol(HDL-C),lowdensity lipoprotein cholesterol(LDL-C)],fasting blood glucose(FBG),fasting insulin(FINS),levels of free fatty acid(FFA),leptin(LP),adiponectin(ADP),visfatin(VF),proliferator-activated receptor γ(PPAR-γ)in adipose tissue,and glucose transporter 4(GLUT-4)in skeletal muscle were determined,and insulin resistance index(IRI)and Lee’s index were calculated. Results Compared with the blank group,the levels of body mass,SBP,DBP,FBG,FINS,TC,TG,LDL-C,FFA,LP,VF,IRI,Lee’s index in the model group were significantly increased,and the levels of HDL-C,ADP,GLUT-4 and PPAR-γ were significantly decreased(all P < 0.05). Compared with the model group,the body mass,SBP,DBP,FBG,FINS,TC,TG,LDL-C,FFA,LP, VF, IRI and Lee’s index in the Chinese medicine group and western medicine group were significantly decreased,and the levels of HDL-C,ADP,GLUT-4 and PPAR-γ were significantly increased(all P < 0.05).Conclusion Modified Linggui Zhugan Decoction has effects on improving glucose and lipid metabolism disorder and insulin resistance in metabolic syndrome rats, whose mechanism may be related with the regulation of adipocytokinessuchasFFA,LP,ADP,VFandPPAR-γ inadiposetissueandGLUT-4 expressioninskeletalmuscle.
