摘要
目的探讨华支睾吸虫感染对FVB和BALB/c小鼠肝纤维化及DX5+NKT细胞的影响。方法解剖麦穗鱼,分离获取华支睾吸虫囊蚴,分别灌胃感染FVB和BALB/c小鼠。无菌分离小鼠肝脏、脾脏并采集外周血,HE和Masson染色法检测两种小鼠肝脏病理变化和纤维化程度,流式细胞术检测外周血淋巴细胞中DX5+NKT细胞的比例。结果华支睾吸虫囊蚴感染4周后,FVB小鼠肝脏和脾脏均明显增大,肝纤维化程度较BALB/c小鼠严重。感染组BALB/c小鼠肝脏中DX5+NKT细胞比例为(5.40±0.40)%,外周血为(3.44±0.60)%,脾脏(2.27±0.20)%,与对照组相比差异均无统计学意义(P>0.05),且在感染1、4.7周后均无明显变化。对照组FVB小鼠的DX5+NKT细胞在肝脏、脾脏和外周血中的水平相似,为1.5%左右。其中肝脏和外周血DX5+NKT细胞比例均显著低于对照组BALB/c小鼠(P<0.05或P<0.01),脾脏DX5+NKT细胞在两种小鼠间差异无统计学意义(P>0.05)。感染4周后,FVB小鼠肝脏DX5+NKT细胞比例为(3.54±0.15)%,对照组为(1.53±0.19)%,差异有统计学意义(P<0.01)。结论BALB/c和FVB小鼠感染华支睾吸虫后DX5+NKT细胞表现出不同的变化和分布,提示其可能在FVB小鼠肝纤维化中发挥作用。
Objective To investigate the effects of a Clonorchis sinensis infection on DX5^+NKT cells and liver fibrosis in mice.Methods C.sinensis metacercariae were collected from Pseudorasbora parva and intragastrically administered to BALB/c and FVB mice.Mice were sacrificed and their livers and spleens were aseptically removed and peripheral blood was collected.Liver injury and fibrosis were assessed using HE and Masson's trichrome staining.The percentage of DX5^+NKT cells in peripheral blood was analyzed using flow cytometry.Results Both the livers and spleens of infected FVB mice were much larger than those of the controls four weeks after infection.Liver injury and fibrosis were much se-vere in FVB mice than in BABL/c mice.The percentage of DX5^+NKT cells was 5.40±0.40%in the liver of infected BALB/c mice,3.44±0.60%in peripheral blood,and 2.27±0.20%in the spleen;percentages did not differ significant-ly from those in the controls even 1,4,and 7 weeks after infection(P>0.05).There were similar percentages(about 1.5%)of DX5^+NKT cells in the liver,spleen,and peripheral blood of control FVB mice.The percentage of DX5^+NKT cells in the liver and peripheral blood of control FVB mice was markedly lower than that in control BALB/c mice(P<0.05 or P<0.01),while the percentage of splenic DX5^+NKT cells did not differ significantly between the two mouse strains(P>0.05).Four weeks after infection,the percentage of DX5^+NKT cells was 3.54±0.15%in the liver of infec-ted FVB mice and 1.53±0.19%in the liver of the control group.The percentage of DX5^+NKT cells differed significant-ly(P<0.01).Conclusion The percentage of DX5^+NKTcells differed in BALB/c and FVB mice after C.sinensis in-fection,indicating that those cells may play a role in fibrosis.
