滋阴补肾方对糖尿病合并去卵巢骨质疏松大鼠病症及骨代谢的影响

Effects of Yin-Nourishing and Kidney-Invigorating Recipe on Signs and Bone Metabolism of Diabetic Rats Complicated with Ovariectomized Osteoporosis

【目的】探讨滋阴补肾方对糖尿病合并去卵巢骨质疏松大鼠病症及骨代谢的影响。【方法】将60只SPF级雌性SD大鼠分为正常组,模型组,骨化三醇组,滋阴补肾方低、中、高剂量组,每组10只。除正常组,其他各组大鼠结扎卵巢,进食高糖高脂饲料并注射链脲佐菌素建立糖尿病骨质疏松模型。造模成功后,对应灌胃给药8周,每日1次。给药结束后,应用生化检测仪检测血糖碱性磷酸酶(ALP)、钙、磷及尿糖水平;酶联免疫吸附分析(ELISA)检测胰岛素、糖化血红蛋白(HbAlc)水平。计算胰岛素抵抗指数。【结果】与正常组比较,模型组的空腹血糖、胰岛素水平及胰岛素抵抗指数显著升高,血糖、尿糖、HbAlc水平显著升高,ALP、钙、磷水平显著降低(均P<0.05);与模型组比较,骨化三醇组和滋阴补肾方低、中、高剂量组的空腹血糖、胰岛素水平及胰岛素抵抗指数显著降低,血糖、尿糖、HbAlc水平显著降低,ALP、钙、磷水平显著升高(均P<0.05),且各治疗组比较,差异均无统计学意义(P>0.05)。【结论】滋阴补肾方可改善糖尿病合并去卵巢骨质疏松大鼠的骨代谢,减轻糖尿病症状,提高胰岛分泌功能。

Objective To explore the effects of Yin-Nourishing and Kidney-Invigorating Recipe(YKR)on signs and bone metabolism of diabetic rats complicated with ovariectomized osteoporosis. Methods Sixty SPF female SD rats were divided into normal group,model group,calcitriol group,and low-,middle-and high-dose YKR groups,10 rats in each group. Apart from the normal group,the rats in the other groups were given ligation of ovaries,feeding with high-sugar and high-fat diet and injection of streptozotocin to establish a diabetic model with ovariectomized osteoporosis. After successful modeling,the rats were respectively given intragastric administration of corresponding drug once a day for 8 weeks. After medication, the contents of blood glucose, alkaline phosphatase(ALP),calcium,phosphorus and urine glucose were detected by biochemical analyzer. The levels of fasting insulin and glycosylated hemoglobin(HbAlc) were detected by enzyme-linked immunosorbent assay(ELISA). The insulin resistance index was calculated. Results Compared with the normal group, the levels of fasting blood glucose,fasting insulin and insulin resistance index were significantly increased in the model group,the levels of blood glucose,urine glucose,HbAlc were significantly enhanced,and the levels of ALP,calcium and phosphorus were significantly decreased(all P<0.05). Compared with the model group,the levels of fasting blood glucose,fasting insulin and insulin resistance index were significantly decreased in the calcitriol group and low-, middle-and high-dose YKR groups, the levels of blood glucose, urine glucose, HbAlc were significantly lowered,and the levels of ALP,calcium and phosphorus were significantly increased(all P<0.05),and there were no differences in above indexes among various treatment group(P>0.05). Conclusion YKR has effects on improving bone metabolism, alleviating diabetic symptoms and improving islet secretion function in diabetic rats with ovariectomized osteoporosis.
