摘要
【目的】探讨蠲痹汤加味对大鼠膝骨性关节炎的影响和作用机制。【方法】将60只SD大鼠随机分为正常组、模型组、蠲痹汤加味组和氨基葡萄糖组,每组15只,通过木瓜蛋白酶注射法建立膝骨性关节炎大鼠模型。造模结束后,蠲痹汤加味组和氨基葡萄糖组分别给予蠲痹汤加味和氨基葡萄糖灌胃,正常组和模型组给予等体积的生理盐水灌胃。灌胃8周后,检测各组大鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素9(IL-9)、一氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD)、转化生长因子β1(TGF-β1)、基质金属蛋白酶3(MMP-3)、组织金属蛋白酶抑制因子1(TIMP-1)的水平,评估关节活动范围和关节滑膜厚度。【结果】与正常组比较,模型组大鼠血清TNF-α、IL-1β、IL-6、IL-9、NO、MDA、MMP-3水平明显升高,SOD、TGF-β1、TIMP-1水平明显降低,关节活动范围减小,关节滑膜厚度增加(均P<0.05);与模型组比较,蠲痹汤加味组大鼠血清TNF-α、IL-1β、IL-6、IL-9、NO、MDA、MMP-3水平明显降低,SOD、TGF-β1、TIMP-1水平明显升高,关节活动范围增加,关节滑膜厚度减小(均P<0.05)。【结论】蠲痹汤加味可能通过抑制炎症反应、氧化应激反应和抑制软骨基质的降解,从而发挥防治KOA的作用。
Objective To explore the effect of modified Juanbi Decoction on knee osteoarthritis in rats and its mechanism. Methods Sixty SD rats were randomly divided into normal group, model group, modified Juanbi Decoction group and glucosamine group, 15 rats in each group. The rat model of knee osteoarthritis was established by papain injection. After modeling,the modified Juanbi Decoction group and glucosamine group were given intragastric administration of modified Juanbi Decoction and glucosamine, respectively, and the normal group and model group were given intragastric administration of normal saline. After 8 weeks of gavage,the serum levels of tumor necrosis factor α(TNF-α),interleukin 1β(IL-1β),interleukin 6(IL-6)and interleukin 9(IL-9),nitric oxide(NO),malondialdehyde(MDA),superoxide dismutase(SOD),transforming growth factor(TGF-β1),matrix metalloproteinase-3(MMP-3),tissue metalloproteinase inhibitor-1(TIMP-1)were detected.The extent of joint motion and synovial thickness of the joint were assessed. Results Compared with the normal group,the serum levels of TNF-α,IL-1β,IL-6,IL-9,NO,MDA and MMP-3 in the model group were significantly increased,the levels of SOD,TGF-β1 and TIMP-1 were significantly decreased,the range of joint activity was decreased,and the thickness of joint synovial membrane was increased(all P<0.05). Compared with the model group, the serum levels of TNF-α, IL-1β, IL-6, IL-9, NO, MDA and MMP-3 of rats were significantly decreased,and SOD,TGF-β1 and TIMP-1 levels were significantly increased in modified Juanbi Decoction group, and the range of joint activity was increased, while the synovial thickness of the joint was reduced(all P<0.05). Conclusion Modified Juanbi Decoction has effects on inhibiting the inflammatory reaction,oxidative stress and degradation of cartilage matrix in rats with knee osteoarthritis.
