加减右归饮对膝骨性关节炎大鼠膝关节软骨退变的影响 被引量:3

摘要 【目的】探讨加减右归饮对膝骨性关节炎模型大鼠膝关节软骨退变的影响。【方法】将120只SD雄性大鼠随机分成假手术组、模型组、加减右归饮组及阳性对照组,每组30只。采用Hulth法将模型组、加减右归饮组及阳性对照组大鼠构建成膝骨性关节炎大鼠模型,假手术组仅打开关节腔。成功造模后,加减右归饮组给予加减右归饮灌胃,阳性对照组给予双醋瑞因(溶于蒸馏水)灌胃,假手术组、模型组给予等量蒸馏水灌胃。给药6周。给药结束后,观察大鼠膝关节影像学表现、血流变学改变、关节液肿瘤坏死因子α(TNF-α)水平、膝关节苏木素-伊红(HE)染色病理学改变及Mankin’s评分。【结果】模型组大鼠影像学可见膝关节退行性改变,全血高切黏度、TNF-α水平均较假手术组明显升高(P<0.01)。加减右归饮组与阳性对照组影像学的膝关节退行性程度、全血高切黏度、TNF-α水平均低于模型组(P<0.01),但仍高于假手术组(P<0.01),而2个治疗组比较,差异均无统计学意义(P>0.05)。【结论】加减右归饮可减轻膝骨性关节炎模型大鼠膝关节软骨退变。 Objective To explore the effects of modified Yougui Yin on degeneration on knee-joint cartilage in rats with knee osteoarthritis. Methods One hundred and twenty SD male rats were randomly divided into shamoperation group,model group,modified Yougui Yin group,and positive control group,30 rats in each group.The rats in the model group, modified Yougui Yin group, and positive control group were induced into knee osteoarthritis model by Hulth method,and the rats in the sham-operation group were only given open knee-joint cavity. After modeling,the modified Yougui Yin group was given intragastric administration of modified Yougui Yin,positive control group was given intragastric administration of diacerein(dissolved in distilled water),and the sham-operation group and model group were given intragastric administration of the same volume of distilled water. The treatment lasted 6 weeks. After medication, the iconographic manifestation of articular cartilage,hemorrheology, tumor necrosis factor α(TNF-α)level in synovial fluid, haematoxylin-eosin(HE)staining pathological features of articular cartilage and Mankin’s scores in rats were observed. Results The iconographic manifestation of degeneration of knee-joint cartilage was seen in the model group,and high blood viscosity and TNF-α level were higher than those in the sham-operation group(P<0.01). The degrees of degeneration of kneejoint cartilage,high blood viscosity and TNF-α level in the modified Yougui Yin group and positive control group were lower than those in the model group(P<0.01),and higher than those in the sham-operation group(P<0.01),there being no significant difference between the two groups(P>0.05). Conclusion Modified Yougui Yin can alleviate degeneration of knee-joint cartilage in rats with knee osteoarthritis.
