嗅三针对帕金森病小鼠α-突触核蛋白及炎症因子MCP-1、HMGB1、IL-1β表达的影响 被引量:12

Effect of Xiusanzhen on the Expressions of α-synuclein and Inflammatory Factors MCP-1,HMGB1 and IL-1β in Mice with Parkinson's Disease

摘要 目的:观察嗅三针对帕金森病(PD)小鼠黑质纹状体区α-突触核蛋白(α-syn)及炎症因子单核细胞趋化蛋白-1(MCP-1)、高迁移率族蛋白1(HMGB1)、白细胞介素-1β(IL-1β)表达的影响,探究嗅三针的作用机制。方法:用随机数字表法将40只健康近交系C57BL/6小鼠分为空白组10只和造模组30只,造模组采用脂多糖(LPS)经鼻滴注造模后分为模型组、电针组、左旋多巴组各10只,电针组对印堂穴和迎春穴进行针刺,左旋多巴组腹腔注射左旋多巴。免疫组化检测黑质纹状体区α-syn表达,Western blotting检测黑质纹状体区炎症因子MCP-1、HMGB1、IL-1β表达。结果:模型组黑质区α-syn及炎症因子MCP-1、HMGB1、IL-1β表达量与空白组比较均上调(P<0.01);电针组黑质区a-syn及炎症因子MCP-1、HMGB1、IL-1β表达量与模型组比较均下调(P<0.05);左旋多巴组黑质区α-syn及炎症因子MCP-1、HMGB1、IL-1β表达量与模型组比较差异无统计学意义(P>0.05)。结论:嗅三针可保护多巴胺能神经元,延缓PD进程,机制可能与其抑制α-syn异常表达及聚集,下调炎症因子表达有关。 Objective:To observe the effect of "xiusanzhen" on the expressions of α-synaptic (α-syn)nuclear protein and inflammatory factors in the substantia nigra striatum of PD mice,and to explore the mechanism of "xiusanzhen". Methods:Total 40 health male C57 BL/6 mice were randomized into the blank group and 3 other groups by using number table method. The three groups were divided into the model group,electroacupuncture group and levodopa group after using nasal drip of LPS,with 10 mice in each group. The protein expression of α-syn in the substantia nigra striatum was detected by immunohistochemistry.The expressions of inflammatory factors MCP-1,HMGB1,IL-1β in the substantia nigra striatum were detected by Western blotting. Results:The expressions of α-syn nucleoprotein and inflammatory factors MCP-1,HMGB1,IL-1β in substantianigra of the model group increased compared with those in the control group(P< 0.01). The expressions of α-syn nucleoprotein and inflammatory factors MCP-1,HMGB1,IL-1β in substantianigra of the electroacupuncture group decreased compared with those in the model group(P< 0.05).The expressions of α-syn nucleoprotein and inflammatory factors MCP-1,HMGB1,IL-1β in substantianigra of the levodopa group were not significantly different from those in the model group(P> 0.05). Conclusions:Xiusanzhen can protect dopaminergic neurons and delay the PD process,which may be related to the inhibition of abnormal expression and aggregation of α-syn nuclear proteins and the reduction of inflammatory factor expression.
