摘要
目的研究miR-155/miR-200b在多囊卵巢综合征(PCOS)中的表达情况及其与PCOS患者临床病理特征的关系,并探讨miR-155/miR-200b在PCOS患者外周血中的作用及意义。方法选取2018年1月-2020年3月杭州市妇产科医院收治确诊为PCOS的患者80例为研究组,另选取同期80例健康非PCOS女性为对照组,回顾性分析两组研究对象临床资料。两组研究对象均进行qRT-PCR检测miR-155、miR-200b表达,研究组患者测定并记录临床病理指标(BMI、超声检查、T、E2、LH/FSH、HOMA-IR),评估miR-155、miR-200b作为PCOS的生物标志物潜能。结果研究组患者血清中miR-155、miR-200b表达水平均高于对照组,差异均有统计学意义(均P<0.05)。BMI> 24 kg/m^2、T>2.5 ng/ml、LH/FSH>3患者miR-155表达量高于BMI≤24 kg/m^2、T≤2.5 ng/ml、LH/FSH≤3患者,差异均有统计学意义(均P<0.05);T>2.5 ng/ml、LH/FSH>3、HOMA-IR>2.69患者miR-200b表达量高于T≤2.5 ng/ml、LH/FSH≤3、HOMA-IR≤2.69患者,差异均有统计学意义(均P<0.05)。miR-155 ROC曲线下面积为0.732,95%CI:0.613~0.801,P<0.001;miR-200b ROC曲线下面积为0.751,95%CI:0.652~0.860,P<0.05。两项指标联合诊断ROC曲线下面积为0.903,95%CI:0.866~0.964,P<0.05。结论 miR-155/miR-200b在PCOS患者血清中高表达,且有望成为诊断PCOS的血清学分子诊断标志。
Objective To study the expression of miR-155/miR-200 b in polycystic ovary syndrome and its relationship with the clinicopathological characteristics of PCOS patients,and to explore the clinical significance of miR-155/miR-200 b. Methods A total of80 patients with PCOS admitted to Hangzhou Obstetrics and Gynecology Hospital from January 2018 to March 2020 as the observation group,and selected 80 healthy non-PCOS patients during the same period as the control group,and retrospectively analyzed their clinical data. Both groups of study subjects performed qRT-PCR to detect the expression of miR-155 and miR-200 b. Patients in the observation group measured and recorded clinicopathological indicators( BMI,ultrasound examination,T,E2,LH/FSH,HOMA-IR). Evaluate the potential of miR-155 and miR-200 b as biomarkers of PCOS. Results The serum levels of miR-155 and miR-200 b in the observation group were higher than those in the control group,and the differences were statistically significant( P<0. 05). The expression level of miR-155 in patients with BMI>24 kg/m^2,T>2. 5 ng/ml,LH/FSH>3 is higher than that of patients with BMI≤24 kg/m^2,T≤2. 5 ng/ml,LH/FSH≤3,the differences were statistically significant( P<0. 05). The expression of miR-200 b in patients with T> 2. 5 ng/ml,LH/FSH> 3,HOMAIR> 2. 69 is higher than that of patients with T≤2. 5 ng/ml,LH/FSH≤3,HOMA-IR≤2. 69,the difference was statistically significant( P<0. 05). The area under the miR-155 ROC curve is 0. 732,95% CI: 0. 613 ~ 0. 801,P<0. 001;miR-200 b ROC area under the curve is 0. 751,95% CI: 0. 652 ~ 0. 860,P<0. 05. The area under the ROC curve for the combined diagnosis of two indicators is 0. 903,95% CI: 0. 866 ~ 0. 964,P<0. 05. Conclusion miR-155/miR-200 b is highly expressed in the serum of PCOS patients,and it is expected to become a serological molecular diagnostic marker for the diagnosis of PCOS.
