TRAIL对骨肉瘤细胞U-2OS凋亡与增殖的影响

摘要 目的探讨TNF相关的凋亡诱导配体(TNF related apoptosis inducing ligand,TRAIL)对骨肉瘤细胞U-2OS凋亡与增殖的影响。方法骨肉瘤U-2OS细胞株随机分为3组,分别为对照组、阿霉素组与联合组,每组(3~5)×105个细胞/ml。对照组进行常规培养,以PBS代替药物处理;阿霉素组采用100μg/ml的阿霉素处理,联合组在阿霉素组处理的基础上采用1000 ng/ml的TRAIL处理。记录处理后不同时间点3组的细胞凋亡、增殖、周期与Caspase-3蛋白表达情况。结果处理后24 h和48 h,相对于对照组,阿霉素组和联合组的细胞增殖指数显著降低(P<0.05),细胞凋亡指数、Caspase-3蛋白相对表达水平显著增加(P<0.05);阿霉素组与联合组的细胞增殖、凋亡指数及Caspase-3蛋白表达水平对比,差异也有统计学意义(P<0.05)。处理后48 h,相比于对照组,阿霉素组与联合组的G 0/G 1期比例显著降低(P<0.05),S期和G 2/M期比例显著增加(P<0.05),阿霉素组与联合组间的对比差异也有统计学意义(P<0.05)。结论TRAIL在骨肉瘤细胞中的应用能促进Caspase-3蛋白的表达,降低G 0/G 1期细胞比例,从而抑制细胞增殖,促进细胞凋亡。 Objective To investigate the effects of TNF-related apoptosis-inducing ligand(TRAIL)on the apoptosis and proliferation of U-2OS in osteosarcoma cell.Methods Osteosarcoma U-2OS cell lines were randomly divided into 3 groups-the control group,the doxorubicin group and the combination group,each group(3~5)×105 cells/ml.The control group was routinely cultured and treated with PBS instead of drugs,the doxorubicin group was treated with 100μg/ml of doxorubicin,and the combined group was treated with 1000 ng/ml of TRAIL on the basis of doxorubicin treatment.The apoptosis,proliferation,cycle and Caspase-3 protein expression were observed in 3 groups.Results At 24 h and 48 h after treatment,the cell proliferation index of the doxorubicin group and the combination group were significantly lower than that of the control group(P<0.05),and the apoptotic index and the relative expression level of Caspase-3 protein were significantly increased(P<0.05),the difference compared between the adriamycin group and the combined group were also statistically significant(P<0.05).At 48 h after treatment,the proportion of G 0/G 1 phase in the combination group was significantly lower than that in the adriamycin group and the combination group(P<0.05),and the proportion of S phase and G 2/M phase were significantly increased(P<0.05),there were also statistically significant difference compared between the 2 groups(P<0.05).Conclusion The application of TRAIL in osteosarcoma cells can promote the expression of Caspase-3 protein and decrease the proportion of G 0/G 1 phase cells,thereby inhibit cell proliferation and promote cell apoptosis.
