藜芦中白藜芦醇提取工艺的优化及对人参黑斑病菌的抑制效果 被引量:2

Optimization of Extraction Technology of Resveratrol From Veratrum nigrum Linn and Its Inhibitory Effect on Alternaria panax

摘要 以藜芦为试材,采用回流、超声波等方法提取藜芦粗粉,并采用高效液相色谱法测定了各个单因素试验中有效成分白藜芦醇含量,根据含量利用响应面分析法寻找最佳提取工艺,进一步采用菌丝生长速率法测定了对人参黑斑病菌的抑菌效果,以期探讨植物藜芦中白藜芦醇提取工艺的优化,并为植物提取物防治人参黑斑病提供参考依据。结果表明:藜芦提取物对人参黑斑病菌的EC50值由小到大依次为甲醇提取物<乙醇提取物<丙酮提取物,藜芦甲醇提取物对人参黑斑病的防治效果优于其它提取物,EC50值为32.04μg·mL^-1;藜芦甲醇提取物的有效成分白藜芦醇最佳提取条件为提取温度59℃,提取时间1.5 h,甲醇体积分数50%,最佳提取量9.33μg·g^-1。 Crude powder of Veratrum nigrum Linn was extracted by refluxing,ultrasonic and other methods,and the content of resveratrol in each single factor test was determined by high performance liquid chromatography.According to the content,response surface analysis was used to find the best extraction process.The antifungal effect of resveratrol on Alternaria panax was further determined by mycelial growth rate method,in order to explore the optimize extraction process of resveratrol from plant V.nigrum Linn,and to provide theoretical basis for the prevention and treatment of A.panax by plant extracts.The results showed that the EC50 value of V.nigrum Linn extract to A.panax was methanol extract<ethanol extract<acetone extract,the control effect of methanol extract of V.nigrum Linn on A.panax was better than that of other extracts,the EC50 value was 32.04μg·mL^-1.The optimum extraction conditions of resveratrol,the effective component of V.nigrum Linn methanol extract,were as follows,extraction temperature 59℃,extraction time 1.5 hours,methanol volume fraction 50%,the optimum extraction amount was 9.33μg·g^-1.
