摘要
对2006-2014年河北省部分养殖场病死鹌鹑采集的气管拭子样品23份,通过病毒分离、血凝试验(HA)、血凝抑制(HI)试验及反转录聚合酶链反应(RT-PCR)鉴定出2株H9N2流感病毒,分别命名为A/Quail/Hebei/LC02/2008(H9N2)(简称Qa/LC02/08)和A/Quail/Hebei/LC01/2011(H9N2)(简称Qa/LC01/11)。通过对HA和NA基因测序后在线BLAST同源性分析表明,Qa/LC02/08与Qa/LC01/11 HA和NA基因的核苷酸同源性分别为97.7%和93.6%;Qa/LC02/08 HA核苷酸与A/Chicken/Shandong/JN/1999(H9N2)及A/Chicken/Zhejiang/ZC28/2007(H9N2)同源性最高,均为99.39%;NA基因核苷酸与A/Chicken/Shandong/N/2010(H9N2)的同源性为99.12%;Qa/LC01/11 HA和NA基因核苷酸与A/Chicken/Hebei/FL/2011(H9N2)同源性分别为99.63%和99.7%。本试验为阐明河北省鹌鹑流感流行病学及制定防控措施提供了科学依据。
The samples of tracheal swabs were collected from dead quails form some farms in Hebei province during 2006-2014.Two strains of H9N2 subtype influenza virus were isolated and identified from 23 tracheal swabs by the viral isolation,hemagglutination(HA)test and hemagglutination inhibition(HI)test,and reverse transcription-polymerase chain reaction(RT-PCR),and were named A/Quail/Hebei/LC02/2008(H9N2)(Abbr.Qa/LC02/08)and A/Quail/Hebei/LC01/2011(H9N2)(Abbr.Qa/LC01/11),respectively.The nucleotide homology of HA gene and NA gene was compared with other strains in NCBI.The homology of HA and NA was 97.7%and 93.6%between Qa/LC02/08 and Qa/LC01/11.The homology of Qa/LC02/08 HA was the highest(99.39%)with A/Chicken/Shandong/JN/1999(H9N2)and A/Chicken/Zhejiang/ZC28/2007(H9N2),and NA was the highest(99.12%)with A/Chicken/Shandong/N/2010(H9N2).The homology of HA and NA of Qa/LC01/11 were 99.63%and 99.7%with A/Chicken/Hebei/FL/2011(H9N2),respectively.The principle was established to provide the epidemiology and prevention and control measures for H9N2 influenza virus from quails in Hebei province.
