褪黑素对帕金森病大鼠模型睡眠障碍的影响 被引量:2

摘要 为研究外源性补充褪黑素是否具有改善帕金森病睡眠障碍的作用及其发挥作用的可能机制。试验将30只大鼠分为正常对照组、帕金森模型组及褪黑素补充组,每组10只。埋置脑电及肌电电极进行多导睡眠记录,高效液相色谱法分析纹状体内神经递质含量变化。结果显示:帕金森模型组总睡眠时间、慢波睡眠时间以及快波睡眠时间与正常对照组相比均明显缩短(P<0.05),觉醒时间则明显延长(P<0.05);褪黑素补充组与帕金森模型组相比总睡眠时间、慢波睡眠时间显著增加(P<0.05),觉醒时间明显减少(P<0.05),两者快波睡眠时间无显著性差异(P>0.05)。褪黑素补充组纹状体内多巴胺含量较帕金森模型组明显增加(P<0.05)。说明外源补充褪黑素可改善帕金森病模型大鼠的睡眠障碍,这种改变可能是通过提高纹状体多巴胺含量来实现。 To verify whether melatonin supplementation can improve sleep disorders in Parkinson’s disease and its possible mechanism.30 rats were divided into normal control group,parkinson model group and melatonin supplementation group with 10 rats in each group.Sleep conductivity was recorded by implanting electroencephalogram(EEG)and electromyogram(EMG)electrodes.The changes of neurotransmitter content in striatum were analyzed by high performance liquid chromatography(HPLC).Compared with the control group,the total sleep time,slow wave sleep time and fast wave sleep time of parkinson model group were significantly shorter(P<0.05),while the wake up time was significantly longer(P<0.05).Compared with parkinson model group,the total sleep time and slow wave sleep time of melatonin supplementation group were significantly increased(P<0.05),while the wake up time was significantly reduced(P<0.05),and there was no difference in fast wave sleep time between them(P>0.05).The content of dopamine in striatum of melatonin supplementation group was significantly higher than that of parkinson model group(P<0.05).The results showed that the exogenous supplementation of melatonin could improve sleep dysfunction in Parkinson’s disease model rats.This change may be achieved by increasing the level of dopamine in the striatum.
