IL-22BP对肠炎相关性结直肠癌小鼠脾单核细胞和结肠组织炎症因子表达的影响

The effect of IL-22BP on the expression of inflammatory factors in spleen monocytes and colon of colitis-associated colorectal cancer mice

目的探讨白细胞介素-22结合蛋白(IL-22BP)对肠炎相关性结直肠癌(colitis-associated colorectal cancer,CAC)小鼠脾单核细胞和结肠组织炎症因子表达的影响。方法建立氧化偶氮甲烷/葡聚糖硫酸钠(AOM/DSS)诱导的肠炎相关性结直肠癌小鼠模型,观察给予白细胞介素-22结合蛋白后小鼠成瘤情况、结肠病理学改变、脾脏和结肠脏器指数,检测脾脏和结肠组织炎症因子干扰素γ(IFN-γ)、白细胞介素-17A(IL-17A)、白细胞介素-22(IL-22)、白细胞介素-6(IL-6)和肿瘤坏死因子α(TNF-α)的变化。结果与模型组相比,白细胞介素-22结合蛋白组小鼠肿瘤数量明显减少,病理检测显示肿瘤评分和组织学活动指数(HAI)评分均显著降低,脾脏指数和结肠指数显著下降,脾单核细胞和结肠组织干扰素γ、白细胞介素-6和肿瘤坏死因子α水平增高,白细胞介素-17A水平降低,白细胞介素-22水平无明显改变。结论白细胞介素-22结合蛋白可以调节肠炎相关性结直肠癌小鼠脾脏和结肠组织炎症因子干扰素γ、白细胞介素-17A、白细胞介素-6和肿瘤坏死因子α的水平。

Objective To investigate the effect of IL-22BP on the expression of inflammatory factors in spleen monocytes and colon tissue of mice with colitis-associated colorectal cancer(CAC).Methods the mice model of CAC was induced by AOM/DSS.The number of tumor,pathological changes,indexes of spleen and colon organs,changes of inflammatory factors IFN-γ,IL-17A,IL-22,IL-6 and TNF-αin spleen and colon were observed.Results IL-22BP reduces AOM/DSS-induced colon tumorigenesis in mice.Compared with the model group,the number of tumor in IL-22BP group was significantly reduced,tumor score and HAI score were significantly reduced,spleen index and colon index were significantly decreased,IFN-γ,IL-6 and TNF-αlevels in spleen monocyte and colon tissue were increased,IL-17A level was decreased,and IL-22 level was not significantly changed.Conclusion IL-22BP can regulate the levels of IFN-γ,IL-17A,IL-6 and TNF-αin spleen and colon of CAC mice.
